In Vivo Characterization of Endogenous Cardiovascular Extracellular Vesicles in Larval and Adult Zebrafish

Summary: We have established a transgenic zebrafish model for in vivo identification, tracking, and extraction of endogenous EVs produced by different cell types. Using a membrane-tethered fluorophore reporter system, we demonstrated that multiple cell types actively produce EVs in zebrafish. High spatiotemporal resolution light-sheet live imaging and modified flow cytometry methods allow for tracking and evaluation of cell-type specific EVs, while cryo electron microscopy reveals the full morphological diversity of larval and adult EVs. This zebrafish model can be utilized for the investigation of endogenous EVs in cardiovascular biology and pathology.
