Chemoselective and Diastereoselective Synthesis of C-Aryl Nucleoside Analogues by Nickel-Catalyzed Cross-Coupling of Furanosyl Acetates with Aryl Iodides

Small RNAs are modified with N-glycans and displayed on the surface of living cells

Summary: This study challenges the conventional view that RNA is not a major target of glycosylation by providing evidence that mammals use RNA as a third scaffold for glycosylation. The researchers found that conserved small noncoding RNAs bear sialylated glycans, which are present in multiple cell types and mammalian species. The majority of glycoRNAs were located on the cell surface and were shown to interact with anti-dsRNA antibodies and members of the Siglec receptor family, suggesting a direct interface between RNA biology and glycobiology.

Synthesis of C-glycosides by Ti-catalyzed stereoselective glycosyl radical functionalization

Mechanistic and computational studies elucidate the unexpected reactivity of glycosyl chlorides in the presence of an organotitanium complex. The utility of this protocol is demonstrated through the concise preparation of therapeutically important carbohydrates and glycopeptide conjugates.

1,2-trans-Stereoselective Synthesis of C-Glycosides of 2-Deoxy-2-amino-sugars Involving Glycosyl Radicals

Nonenzymatic Stereoselective S-Glycosylation of Polypeptides and Proteins

The method was successfully applied for direct glycosylation of complex polypeptides and proteins after developing a second-generation protocol, with computational studies performed to elucidate the reaction mechanism.

Light-Mediated Cross-Coupling of Anomeric Trifluoroborates

Summary: C-Glycosyl peptides/proteins, stable mimics of native glycopeptides/proteins with therapeutic potential, have been successfully synthesized using a novel method. This synthesis approach is versatile, accommodating a wide range of coupling partners, including complex oligosaccharides and peptide substrates. The resulting vinyl C-glycosyl amino acids and peptides are amenable to further transformations for elongation of peptide and saccharide chains.

Photoredox-Catalyzed Stereoselective Synthesis of C-Nucleoside Analogues from Glycosyl Bromides and Heteroarenes

Summary: The synthesis of C-nucleoside analogues through photoredox-catalyzed radical coupling allows for good functional group tolerance and site- and stereoselectivities, making it applicable for coupling various monosaccharides, disaccharides, and polysaccharides with nonfunctionalized heteroarenes. Biologically important molecules can be glycosylated directly through a C-C bond using this method, and the site- and stereoselectivities can be rationalized and predicted by density functional theory (DFT) calculations.
