In Vitro Flow Chamber Design for the Study of Endothelial Cell (Patho)Physiology

Meghan E. Fallon et al.

Summary: Flowing blood produces frictional force on blood vessel walls, which affects endothelial cell function and phenotype. Shear stress in the flow environment plays a role in the pathogenesis of atherosclerosis and cardiovascular diseases. Accurately simulating in vivo hemodynamics is crucial for studying endothelial cell responses.

Low shear stress regulates vascular endothelial cell pyroptosis through miR-181b-5p/STAT-3 axis

Xiangshan Xu et al.

Summary: The study revealed that low shear stress can suppress the expression of mechanosensitive miR-181b-5p in HUVECs, leading to an increase in NLRP3 inflammasome-dependent pyroptosis. Inhibition of STAT-3 gene expression at the posttranscriptional level can alleviate NLRP3 inflammasome-dependent pyroptosis. Regulation of the expression levels of mechanosensitive miR-181b-5p and STAT-3 may be a potential strategy to control atherosclerosis induced by shear stress.

Estrogen prevent atherosclerosis by attenuating endothelial cell pyroptosis via activation of estrogen receptor α-mediated autophagy

Qinghai Meng et al.

Summary: The study found that post-menopausal women have decreased autophagy and ER alpha expression in the aortic endothelium, with increased inflammation and pyroptosis. Estrogen treatment can accelerate autophagy and improve cell pyroptosis in OVX ApoE-/- mice and Hcy-treated HUVECs. Estrogen prevents atherosclerosis by upregulating ERα expression and subsequently inducing autophagy to reduce inflammation and pyroptosis.

Rac1/Wave2/Arp3 Pathway Mediates Rat Blood-Brain Barrier Dysfunction under Simulated Microgravity Based on Proteomics Strategy

Ranran Yan et al.

Summary: This study investigated the impact of simulated microgravity (MG) on the blood-brain barrier (BBB) in rats and human brain microvascular endothelial cells. The results showed that MG could disrupt the BBB by affecting oxidative stress levels, proinflammatory cytokine levels, cell structure, and junction protein expression. The study also identified the potential involvement of the Rac1/Wave2/Arp3 signaling pathway in BBB dysfunction induced by MG.

Substrate stiffness differentially impacts autophagy of endothelial cells and smooth muscle cells

Mi Hu et al.

Summary: The stiffness of the vascular extracellular matrix plays a crucial role in artery pathology and can impact the autophagy levels of vascular endothelial cells and smooth muscle cells. Increasing substrate stiffness leads to differential changes in autophagy levels, affecting gene expression associated with endothelial function and transitioning VSMCs from contractile to synthetic phenotype. Inhibiting cell autophagy further reduces endothelial functional gene expression and increases VSMC calponin expression, highlighting the important role of autophagy in response to changes in microenvironment stiffness.

Summary: ER stress resulting from ischemic injury initiates unfolded protein response (UPR) as a protective mechanism to maintain ER homeostasis. Mild ER stress helps cells to escape from danger signals and activate pro-survival mechanisms to rescue ischemic injury, while chronic ER stress can trigger various pro-apoptotic mechanisms, leading to detrimental effects on nerve cells.
