Loss of hepatic Mboat7 leads to liver fibrosis

Veera Raghavan Thangapandi et al.

Summary: The rs641738C>T variant near the MBOAT7 locus is associated with liver fibrosis in various liver diseases. Mice with Mboat7 deficiency and human carriers of rs641738TT with fibrosis showed alterations in lipid signaling, suggesting a potential targetable treatment option for NAFLD.

rs641738C>T near MBOAT7 is associated with liver fat, ALT and fibrosis in NAFLD: A meta-analysis

Kevin Teo et al.

Summary: The genetic variant rs641738C>T near MBOAT7 is validated as a risk factor for the presence and severity of non-alcoholic fatty liver disease (NAFLD) in individuals of European descent. This variant is associated with higher liver fat, NAFLD diagnosis, and advanced fibrosis in Caucasian adults.

Hepatic deletion of Mboat7 (LPIAT1) causes activation of SREBP-1c and fatty liver

Mingfeng Xia et al.

Summary: Recent identification of genetic variants that increase the risk of fatty liver disease and cirrhosis has led to the characterization of Mboat7 liver-specific KO mice in order to elucidate the link between these variants and fatty liver disease. The study revealed a relationship between fatty acid composition and regulation of hepatic fat synthesis, as well as delineated the mechanism by which mutations in MBOAT7 cause hepatic steatosis. Hepatic removal of both SREBP cleavage-activating protein (Scap) and Mboat7 normalized hepatic triglycerides relative to Scap-only hepatic KO, showing that increased SREBP-1c processing is required for Mboat7-induced steatosis.

Summary: Phosphatidylcholine (PC) undergoes re-acylation catalyzed by lysophosphatidylcholine acyltransferases (LPCAT), with LPCAT3 introducing polyunsaturated acyl to modulate membrane fluidity and membrane protein functions. The structures of LPCAT3 reveal a reaction chamber and tunnels for substrate positioning, providing insights into the re-acylation mechanism.

Photochemical Probe Identification of a Small-Molecule Inhibitor Binding Site in Hedgehog Acyltransferase (HHAT)**

Thomas Lanyon-Hogg et al.

Summary: This study reports the rational development of a photochemical probe to investigate a novel small-molecule inhibitor binding site in the human MBOAT Hedgehog acyltransferase (HHAT), leading to the identification of the most potent HHAT inhibitor IMP-1575 to date. Through structure-activity relationship investigation and methods such as photocrosslinking, the study revealed the inhibition mechanism and small molecule binding site of HHAT, providing a strategy for mapping small molecule interaction sites in MBOATs.

LPIAT1/MBOAT7 contains a catalytic dyad transferring polyunsaturated fatty acids to lysophosphatidylinositol

Andrea Caddeo et al.

Summary: MBOAT7 catalyzes the transfer of PUFAs to lipid acceptors and shows the highest affinity for LPI, while missense mutations at the MBOAT7 putative catalytic dyad inhibit the O-acyltransferase activity of the protein. The association between the MBOAT7 rs641738 variant and the increased risk of NAFLD is mediated by changes in the hepatic phosphatidylinositol acyl-chain remodeling.
