A method for assaying peptide: N-glycanase/N-glycanase 1 activities in crude extracts using an N-glycosylated cyclopeptide

Hiroto Hirayama et al.

Summary: The deficiency of cytosolic peptide N-glycanase (NGLY1) is a rare genetic disorder with common symptoms exhibited by patients, however, there is currently a lack of a reliable method to quantify NGLY1 activity.

Cytosolic Free N-Glycans Are Retro-Transported Into the Endoplasmic Reticulum in Plant Cells

Makoto Katsube et al.

Summary: The study found that high-mannose type N-glycans produced in plant cells have inhibitory effects on protein folding and beta-amyloid fibril formation, while there are also fully processed complex type N-glycans present in the Golgi apparatus. These FNGs may be transported back to the ER and processed in the Golgi apparatus through the protein secretion pathway.

JF1/B6F1 Ngly1-/- mouse as an isogenic animal model of NGLY1 deficiency

Makoto Asahina et al.

Summary: This study successfully created viable NGLY1-deficient mice by crossbreeding different mouse strains. These mice exhibited developmental delay and motor dysfunction similar to human patients, making them a valuable model for preclinical testing and understanding the pathogenic mechanisms of NGLY1 deficiency.

Loss of peptide:N-glycanase causes proteasome dysfunction mediated by a sugar-recognizing ubiquitin ligase

Yukiko Yoshida et al.

Summary: Mutation in the NGLY1 gene leads to a recessive disorder in humans. Findings in this study suggest that dysfunction of the proteasome caused by accumulation of N-glycoproteins, particularly NRF1 ubiquitinated by SCFFBS2, contributes to the pathogenesis resulting from NGLY1 deficiency.

Reversibility of motor dysfunction in the rat model of NGLY1 deficiency

Makoto Asahina et al.

Summary: NGLY1 deficiency is a rare inherited disorder with no current effective therapies, with Ngly1-/- rat model showing similar symptoms. Treatment with AAV9-hNGLY1 can restore NGLY1 expression and correct motor phenotypes, suggesting the CNS as a primary therapeutic target for NGLY1 deficiency.
