Summary: Our study demonstrates that the gut microbiota-inosine-A(2A)R/PPAR gamma axis is crucial in maintaining intestinal homeostasis and may offer a novel approach for preventing colitis through manipulation of gut microbial purine metabolite. Additionally, dietary supplementation of barley leaf improves intestinal mucosal barrier functions and induces metabolic reprogramming in colonic tissues by enhancing the glycolysis process. Furthermore, the enrichment of microbiota-derived purine metabolite inosine from BL supplementation activates PPAR gamma signaling in human colon epithelial cells.
