Surface charge, glycocalyx, and blood-brain barrier function

Summary: The negative surface charge of brain microvessel endothelial cells is derived from the special composition of their membrane lipids and the thick endothelial surface glycocalyx. This charge plays a crucial role in the permeability of macromolecules and nanoparticles as well as drug interactions. Changes in surface charge and glycocalyx in brain microvasculature-related pathologies may require protective measures to prevent damage.

Modified cyclodextrin-based nanoparticles mediated delivery of siRNA for huntingtin gene silencing across an in vitro BBB model

Summary: Researchers have developed a novel delivery system based on modified cyclodextrin nanoparticles (CDs) loaded with small interfering RNAs (siRNAs) targeting HTT and complexed with the rabies virus glycoprotein (RVG). This system successfully crosses the brain endothelial cells, releases the encapsulated siRNAs into the cytoplasm of neuronal cells, and mediates downregulation of HTT. The CD platform shows promise as an option for delivering siRNA-based therapeutics for HD and potentially other diseases with genetically validated targets in the central nervous system.

Gene Therapy for Polyglutamine Spinocerebellar Ataxias: Advances, Challenges, and Perspectives

Summary: Polyglutamine spinocerebellar ataxias are a group of autosomal dominant disorders caused by repeat expansions in single genes. While there is currently no curative treatment available, gene therapy strategies like RNA interference and antisense oligonucleotide therapy have shown promise in cellular and animal models. Gene editing strategies such as CRISPR/Cas9 have also been introduced to a limited extent, but face technological and ethical challenges in their potential use in future clinical trials.

Gene therapy for ALS: A review

Summary: ALS has historically been challenging for gene therapy due to a lack of therapeutic targets and difficulty accessing the brain and spinal cord. Recent advances in disease mechanism understanding, ALS genetics, and CNS targeting, along with progress in gene delivery and editing techniques, have opened new therapeutic possibilities. Gene therapy clinical trials are underway for ALS patients with various genetic mutations, showing potential for disease-modifying treatment.

Tissue distribution and cellular localization of gold nanocarriers with bound oligonucleotides

Summary: The aim of the study was to investigate the effect of DNA cargo on tissue distribution of gold glyconanoparticles, and the results showed that the addition of DNA changed the localization of the nanoparticles in liver hepatocytes and splenic macrophages, but not in heart, lung, or brain. DNA increased the size and added a strong negative charge to the nanoparticles, which significantly affected tissue distribution.

Summary: This study demonstrates that DNA/RNA heteroduplex oligonucleotides (HDOs) conjugated to cholesterol or alpha-tocopherol can regulate endogenous gene expression in the central nervous system when administered systemically, with limited side effects. The HDOs effectively distribute throughout the CNS and suppress the expression of target genes by up to 90%, showing potential for ASO-based therapies for neurological diseases.

Comparison of polypeptides that bind the transferrin receptor for targeting gold nanocarriers

This study compared different polypeptides for their ability to bind to brain endothelium and target therapeutic agents to the CNS. The findings showed that attaching a TfR-targeting polypeptide significantly increased endocytosis rates by brain endothelium and enhanced nanoparticle movement across cells.
