The antidiabetic drug glibenclamide exerts direct retinal neuroprotection

Marianne Berdugo et al.

Summary: Sulfonylureas, commonly used for hypoglycemia in adults with type 2 diabetes, have shown neuroprotective effects in central nervous system injury models and children with neuropsychomotor impairments from neonatal diabetes. Glibenclamide, a sulfonylurea, has been found to protect retinal structure and function in various retinal injury models, indicating potential for repurposing in diabetic retinopathy treatment.

Cerebrospinal Fluid Sulfonylurea Receptor-1 Is Associated with Intracranial Pressure and Outcome after Pediatric TBI: An Exploratory Analysis of the Cool Kids Trial

Benjamin E. Zusman et al.

Summary: The study found that sulfonylurea receptor-1 (SUR1) is increased in pediatric severe traumatic brain injury (TBI) and is associated with intracranial pressure and outcome, in contrast to findings in adults. This suggests that SUR1 may be a potential therapeutic target in pediatric TBI, warranting further investigation.

The effects of trifluoperazine on brain edema, aquaporin-4 expression and metabolic markers during the acute phase of stroke using photothrombotic mouse model

Nicole J. Sylvain et al.

Summary: Stroke is the second leading cause of death and the third leading cause of disability worldwide, with edema playing a major role in stroke-associated morbidity and mortality. Regulating water balance in the brain through targeting AQP4 channels may be a promising therapeutic approach for treating brain edema. The FDA-approved drug trifluoperazine (TFP) has shown efficacy in reducing cerebral edema in post-stroke mice by inhibiting AQP4 expression, without inducing deleterious changes in brain electrolytes or metabolic markers.

Abcc8 (Sulfonylurea Receptor-1) Impact on Brain Atrophy after Traumatic Brain Injury Varies by Sex

Swathi Tata et al.

Summary: The research suggests that Abcc8 knockout mice exhibit smaller contusion volumes and larger normalized contralateral hemisphere volumes after controlled cortical impact, with these effects being more significant in male mice. Additionally, in vitro studies indicate that the sex-determining region Y (SRY) may play a crucial role in these outcomes.

Molecular mechanisms of brain water transport

Nanna MacAulay

Summary: Our brains consist of 80% water, which is continuously shifted between different compartments and cell types during physiological and pathophysiological processes. Disturbances in brain water homeostasis occur with pathologies such as brain oedema and hydrocephalus. Although aquaporins greatly accelerate the movement of water, they do not suffice to explain overall brain water movement, suggesting the presence of unknown molecular mechanisms.

The inclusion of genetic variation in clinical models improved prediction accuracy, suggesting potential implications for precision medicine in severe TBI.
