Toxicological Analysis of Cases of Mixed Poisonings with Synthetic Cathinones and Other Drugs of Abuse

Ewelina Pieprzyca et al.

Summary: The use of synthetic cathinones (SCs) poses a high risk to human health and life and has a significant addictive potential. Most SC users take multiple substances simultaneously, including traditional drugs and other new psychoactive substances. This study provides pioneering data on the concentrations and effects of different SCs, as well as the presence of other substances in biological samples. Taking SCs can have serious side effects, leading to organ failure and death. Additionally, the combined use of multiple psychoactive substances increases the toxicity of SCs.

Summary: Synthetic cathinones have been increasingly popular since 2003 and have become the most seized class of new psychoactive substances reported to the UNODC. Recent studies on SC-related fatalities globally have highlighted new consumption trends and symptoms. Analytical methods and mass spectrometry instruments play a crucial role in detecting the low concentrations of these potent new SCs.

Role of amino terminal substitutions in the pharmacological, rewarding and psychostimulant profiles of novel synthetic cathinones

L. Duart-Castells et al.

Summary: The study highlights the role of amino terminal substitution in the pharmacological profile of novel synthetic cathinones, including their potency in inhibiting dopamine uptake and inducing psychostimulant and rewarding effects in mice. The results demonstrate a significant correlation between predicted binding affinities by molecular docking and affinity constants for human dopamine transporter, indicating the importance of amino substitutions in determining the biological activity of these compounds.

The acute effects of multiple doses of methamphetamine on locomotor activity and anxiety-like behavior in adolescent and adult mice

Hayley A. Ortman et al.

Summary: The effects of MA on adolescent and adult mice are not affected by age and dose, but adolescent mice show a stronger locomotor response to high dose MA while adult mice show a stronger response to low dose MA. There is no difference in plasma corticosterone levels between the two age groups exposed to MA. Further research is needed to understand the developmental neurobiological mechanisms underlying the age differences in response to acute MA exposure.

Structure-Activity Relationship of Novel Second-Generation Synthetic Cathinones: Mechanism of Action, Locomotion, Reward, and Immediate-Early Genes

Nuria Nadal-Gratacos et al.

Summary: The study investigated the structure-activity relationships of six novel synthetic cathinones, revealing potent dopamine uptake inhibition and varying effects on serotonin transporters. These synthetic cathinones exhibited different psychostimulant effects and rewarding properties in mice, providing valuable information for understanding their mechanisms of action in addiction.

Summary: This case study presents a fatal poisoning involving several cathinones, detected and quantified using LC-ESI-LIT-MS in authentic human whole blood samples. The LC-ESI-LIT-MS method showed high specificity for qualitative analysis of cathinones compared to GC-EI-MS. Validation data revealed good linearity and reproducibility for quantitative analysis of the compounds.
