Leber congenital amaurosis/early-onset severe retinal dystrophy: current management and clinical trials

Malena Daich Varela et al.

Summary: Current research focuses on genetic therapies for Leber congenital amaurosis (LCA) and various clinical trials are ongoing globally to achieve better outcomes. Gene therapy, cell therapy, and pharmacological approaches show potential for the treatment of LCA, with advancements in techniques like CRISPR-mediated gene editing and antisense oligonucleotides providing new possibilities.

Functional Characterization of Rare Genetic Variants in the N-Terminus of Complement Factor H in aHUS, C3G, and AMD

Edwin K. S. Wong et al.

Summary: MPGN, C3G, aHUS, and AMD are closely linked to dysfunction of the AP, with rare genetic variants in the CFH gene causing deficiencies in the FH protein. Careful biochemical assessment of these variants is crucial for accurate clinical decisions and patient management.

Predicting quality of life in AMD patients-insights on the new NICE classification and on a bolt-on vision dimension for the EQ-5D

Ricardo De Sousa Peixoto et al.

Summary: The study found that visual dysfunction in AMD significantly affects HRQoL and handicap. EQ-5D was less sensitive compared to VF-14 and LHS, and not significantly correlated with the VIS bolt-on domain. The new AMD classification was a good predictor of visual HRQoL and had an excellent association with visual acuity.

Retinal Inflammation, Cell Death and Inherited Retinal Dystrophies

Lorena Olivares-Gonzalez et al.

Summary: Inherited retinal dystrophies (IRDs) are a group of retinal disorders that lead to progressive vision loss due to retinal cell death, predominantly photoreceptor cells. The treatment of IRDs is complicated by genetic and clinical heterogeneity, but reducing retinal inflammation has emerged as an effective approach to slowing disease progression.

LEBER CONGENITAL AMAUROSIS DUE TO CEP290 MUTATIONS-SEVERE VISION IMPAIRMENT WITH A HIGH UNMET MEDICAL NEED A Review

Bart P. Leroy et al.

Summary: LCA10, caused by CEP290 mutations, is a genetic retinal disease leading to severe visual impairment in early childhood. There are currently no approved treatments, but investigational strategies include RNA editing using antisense oligonucleotides or CRISPR-based approaches. The antisense oligonucleotide therapy QR-110 has shown promising results in early trials, with ongoing phase 3 clinical trials.

NLRP3 Inflammasome at the Interface of Inflammation, Endothelial Dysfunction, and Type 2 Diabetes

Ilona M. Gora et al.

Summary: Type 2 diabetes mellitus is characterized by chronic inflammation, and the activation of the NLRP3 inflammasome plays a significant role in this condition. Endothelial dysfunction is identified as a key step in inflammasome activation, suggesting that targeting the NLRP3 inflammasome could offer new therapeutic approaches.

Mesenchymal Stem Cell-Based Therapy for Retinal Degenerative Diseases: Experimental Models and Clinical Trials

Vladimir Holan et al.

Summary: Cell therapy using mesenchymal stem cells (MSCs) offers a potential treatment option for retinal degenerative diseases by utilizing the cells' immunoregulatory and anti-inflammatory properties to support retinal cell survival and growth. MSCs also have the potential to protect retinal cells and contribute to the regeneration of the diseased retina.

Molecular Mechanisms Related to Oxidative Stress in Retinitis Pigmentosa

Carla Enrica Gallenga et al.

Summary: Retinitis pigmentosa is a genetic retinopathy, but non-genetic factors such as inflammation, autophagy, and oxidative stress play a crucial role in its pathogenesis and progression. Oxidative stress, caused by constant exposure to light/oxidative stress, results in accumulation of ROS in the retina, leading to microglial activation and photoreceptor degeneration. Various genetic mutations and alterations in cellular response contribute to the vicious cycle of neuroinflammation and degeneration in RP, suggesting microglia as a key target for oxidative stress therapy.
