Small-molecule CD73 inhibitors for the immunotherapy of cancer: a patent and literature review (2017-present)

Summary: Hydrolysis of AMP to adenosine and inorganic phosphate is catalyzed by the metalloenzyme 5'-ectonucleotidase (e5NT), also known as CD73, which has two zinc ions at its active site. CD73 inhibitors, including monoclonal antibodies (MAbs) and small molecules, are being considered for tumor immunotherapy. Considerable progress has been made in the design of nucleotide/nucleoside-based CD73 inhibitors, with a highly potent small-molecule inhibitor called AB680 currently in early clinical trials.
