Genome structural variation in human evolution

Summary: Structural variation (SV) refers to large differences in genomic structure between two genomes, including both copy number variations and variations that do not change copy number, such as inversions. Recent advances in reference genomes and sequencing technologies have renewed interest in SV research, highlighting its role in human evolution. Studies have shown the impact of SV on changes in human brain, population-specific responses to the environment, and contributions from archaic hominin admixture to present-day human SV.

Long-read sequencing settings for efficient structural variation detection based on comprehensive evaluation

Summary: With the rapid development of long-read sequencing technologies, it is now possible to reveal the full spectrum of genetic structural variation (SV). However, the high cost, limited read length, and sequencing errors associated with long-read data have hindered the widespread adoption of SV calling. This study provides important guidelines for selecting long-read sequencing settings for efficient SV calling, with high sequencing coverage being the most influential factor in promoting SV calling.

Benchmarking germline CNV calling tools from exome sequencing data

Summary: The study aimed to comprehensively analyze tools capable of germline CNV calling using a single CNV standard and reference sample set. Results showed that while most tools could detect a range of CNV lengths, they exhibited low concordance with each other. After unified comparison, it was found that the tools were not equivalent, allowing for the selection of algorithms or ensembles of algorithms most suitable for specific goals.
