Summary: The issue of plastic waste has become a major topic on the international agenda due to the inability to effectively manage the increasing quantity of plastic materials produced. Various initiatives, including legal regulations, have been introduced to reduce plastic waste, but decisions to limit production or use of plastics are inevitably influenced by economic evaluations and the shortcomings of alternative materials.

QM/MM Study of the Enzymatic Biodegradation Mechanism of Polyethylene Terephthalate

Sergio Boneta et al.

Summary: This study investigated the enzymatic biodegradation of PET at an atomic level using molecular dynamics simulations. Two hydrolases, Ideonella sakaiensis 201-F6 and leaf-branch compost cutinase, showed degradation activity towards PET. The study revealed a characteristic four-step degradation pathway for both enzymes, with LCC-ICCG variant outperforming PETase possibly due to its ability to work at higher temperatures and its intrinsic relationship with the crystallinity grade of the polymer.

Assessment of the PETase conformational changes induced by poly(ethylene terephthalate) binding

Clauber Henrique Souza da Costa et al.

Summary: A bacterium strain with the ability to degrade PET was identified, and its enzyme IsPETase catalyzes the hydrolysis of PET. Molecular docking and dynamics simulations were used to analyze structural changes of IsPETase induced by PET binding, suggesting strategies for increasing the enzyme's thermal stability. This study provides insights into the molecular binding mechanism of PET into IsPETase structure and a computational strategy for mapping flexible regions of the enzyme.
