Temozolomide: An Updated Overview of Resistance Mechanisms, Nanotechnology Advances and Clinical Applications

Summary: Temozolomide is an oral alkylating prodrug commonly used in the treatment of high-grade gliomas, with high oral bioavailability and the ability to cross the blood-brain barrier. However, despite its efficacy, patient survival remains relatively short, largely due to tumor drug resistance. Patterns of glioblastoma treatment and survival over a 16-years period: pooled data from the German Cancer Registries

Summary: This study analyzed 40,138 adult GBM cases diagnosed in Germany between 1999 and 2014, finding that younger age, female sex, surgical resection, and the use of radiotherapy and chemotherapy were associated with better survival outcomes. The inclusion of temozolomide chemotherapy significantly improved median OS in older age groups but had a lesser effect in younger age groups. Survival improvements were observed over time for each treatment option.
