Structural, functional, and mechanistic insights uncover the fundamental role of orphan connexin-62 in platelets

Khaled A. Sahli et al.

Summary: Connexins form hexameric hemichannels in the plasma membrane, which can dock together to form gap junctions and facilitate intercellular trafficking of molecules. A study on the orphan connexin Cx62 in human and mouse platelets revealed the development of a novel mimetic peptide ((62)Gap27) to interfere with its function and its impact on platelet functions. The inhibition of Cx62 by (62)Gap27 suppressed platelet functions and impaired thrombosis, showing a fundamental role of Cx62 in regulating circulating cell functions.

Danegaptide Prevents TGFβ1-Induced Damage in Human Proximal Tubule Epithelial Cells of the Kidney

Paul E. Squires et al.

Summary: Danegaptide has been shown to block hemichannel-mediated changes in protein expression and function in tubular epithelial kidney cells, protecting against tubular injury. It also inhibits TGF beta 1-induced changes in the expression and secretion of key adipokines, cytokines, chemokines, growth factors, and interleukins, indicating its potential in the future treatment of CKD.

Cx43 hemichannel microdomain signaling at the intercalated disc enhances cardiac excitability

Maarten A. J. De Smet et al.

Summary: Cx43 hemichannels are activated during diastolic Ca2+ release in ventricular cardiomyocytes, contributing to enhanced Ca2+ dynamics due to Ca2+ entry and coupling to release microdomains. Hemichannel opening also leads to delayed afterdepolarizations and triggered action potentials. Increased hemichannel activity is associated with electrical instability in failing human hearts compared to nonfailing donor hearts, suggesting a potentially novel mechanism of cardiac arrhythmogenesis in heart failure.

Activation of Cx43 Hemichannels Induces the Generation of Ca2+ Oscillations in White Adipocytes and Stimulates Lipolysis

Egor A. Turovsky et al.

Summary: The study investigated the mechanisms of Ca2+ oscillation generation and regulation of the lipolysis/lipogenesis balance in white adipocytes in response to a decrease in extracellular calcium concentration. It was found that activation of connexin-43 led to ATP release from adipocytes, causing changes in gene expression related to lipolysis and a decrease in lipid droplet accumulation.

Peptidic Connexin43 Therapeutics in Cardiac Reparative Medicine

Spencer R. Marsh et al.

Summary: Channels formed by Connexin (Cx43) are associated with various heart diseases, with some Cx43 mimetic peptides showing therapeutic potential and advancing to clinical testing. However, challenges in drug delivery, stability, and tissue penetration have caused reluctance in the pharmaceutical industry to translate peptidic therapeutics to the clinic. Novel drug delivery technologies like nanoparticles may provide solutions to these issues.

Summary: Compounds that block the function of connexin and pannexin protein channels have shown potential as valuable therapeutics for diseases, but the current literature on their molecular effects on tissues is inconclusive. This review discusses the distinct physiological and pathological implications of blocking different channel types, as well as the contribution of connexins and pannexins to excessive ATP release linked to disease onset and progression.
