Establishment of appropriate glaucoma models using dexamethasone or TGFβ2 treated three-dimension (3D) cultured human trabecular meshwork (HTM) cells

Megumi Watanabe et al.

Summary: By culturing human trabecular meshwork cells in the presence of dexamethasone or TGF beta 2, researchers were able to establish suitable ex vivo models for studying steroid-induced and primary open-angle glaucoma. These models showed changes in extracellular matrix expression and spheroid size and stiffness, making them valuable tools for glaucoma research.

Prostaglandin F2α agonists induced enhancement in collagen1 expression is involved in the pathogenesis of the deepening of upper eyelid sulcus

Kaku Itoh et al.

Summary: The study reveals that PGF2α-ags can significantly reduce the size of 3D organoids and increase their physical solidity in human orbital fibroblasts. This effect is concentration-dependent and can impact the mRNA expression of genes related to adipogenesis and extracellular matrix remodeling.

The Expression Regulation and Biological Function of Autotaxin

Xiaotian Zhang et al.

Summary: Autotaxin plays a crucial role in cancer development by promoting cell proliferation, migration, and survival through producing LPA, as well as increasing the expression of inflammation-related circuits that impact patients' prognosis. ATX is considered a potential therapeutic target for cancer, with a number of ATX inhibitors currently developed.

Diverse effects of pan-ROCK and ROCK2 inhibitors on 2 D and 3D cultured human trabecular meshwork (HTM) cells treated with TGFβ2

Megumi Watanabe et al.

Summary: This study investigated the effects of ROCK1 and ROCK2 on TGF beta 2-treated HTM cells using a pan-ROCK inhibitor and a ROCK2 inhibitor. The inhibitors were found to modulate the expression of extracellular matrix and affect cell function. TGF beta 2 treatment resulted in changes in cell function and gene expression, which were significantly and differently modulated by the inhibitors.

Screening of the Drug-Induced Effects of Prostaglandin EP2 and FP Agonists on 3D Cultures of Dexamethasone-Treated Human Trabecular Meshwork Cells

Megumi Watanabe et al.

Summary: This study aimed to screen the drug-induced effects of PGF2α and OMD on DEX-treated HTM cells using 2D and 3D cell cultures, revealing that PGF2α and OMD could inhibit some adverse effects induced by DEX and affect the gene expression of the cells.

Crosstalk between transforming growth factor β-2 and Autotaxin in trabecular meshwork and different subtypes of glaucoma

Nozomi Igarashi et al.

Summary: The TGF-β2/ATX ratio was significantly higher in AH of control or POAG compared with SOAG, and negatively correlated with IOP. ATX expressions were regulated by TGF-β2 in hTM cells in a concentration-dependent manner. This study suggests that trans-signaling of TGF-β2 plays a role in regulating ATX expressions and inducing upregulations of TGF-βs or fibrosis in hTM cells, reflecting different profiles of these mediators in glaucoma subtypes.
