Catalytic activity profile of polyP:AMP phosphotransferase from Myxococcus xanthus

Yoshio Kimura et al.

Summary: Myxococcus xanthus generates polyphosphates during starvation and forms fruiting bodies through the activity of Pap. Pap requires higher concentrations of Mn2+ or Mg2+ ions for longer polyPs to demonstrate maximum activity, with a lower K-m for longer polyPs and the highest k(cat) for polyP(60-70). This suggests a functional role of Pap during M. xanthus starvation in generating ATP from AMP, ADP, and polyP.

Broad-Spectrum Inhibitor of Bacterial Polyphosphate Homeostasis Attenuates Virulence Factors and Helps Reveal Novel Physiology of Klebsiella pneumoniae and Acinetobacter baumannii

Nathan Roberge et al.

Summary: This study demonstrated that the small molecule gallein, which exhibits broad-spectrum PPK inhibition, successfully attenuated virulence factors in multiple antibiotic-resistant pathogens, particularly in A. baumannii. These findings suggest that gallein has the potential to be developed into a broad-spectrum anti-virulence therapeutic option.

Recent Developments and Challenges for the Industrial Implementation of Polyphosphate Kinases

Michele Tavanti et al.

Summary: Polyphosphate kinases (PPKs) have emerged as a promising solution for the scalable recycling of ATP, using polyP salts as the phosphate donor. This review discusses recent efforts in characterizing PPKs and the challenges associated with implementing them in chemical manufacturing, identifying gaps that need to be addressed for successful adoption in industry. Guidelines for a wider adoption of PPKs are also provided.

A Dual-Specificity Inhibitor Targets Polyphosphate Kinase 1 and 2 Enzymes To Attenuate Virulence of Pseudomonas aeruginosa

Nolan Neville et al.

Summary: The small molecule gallein was discovered as a dual-specificity inhibitor of both PPK1 and PPK2 enzyme families in Pseudomonas aeruginosa, disrupting cellular polyphosphate accumulation and attenuating biofilm formation, motility, and toxin production. This study establishes PPK2 enzymes as valuable drug targets in P. aeruginosa and provides a promising starting point for the development of new antivirulence therapeutics.

Targeting Polyphosphate Kinases in the Fight against Pseudomonas aeruginosa

Kanchi Baijal et al.

Summary: Polyphosphate (polyP) is a conserved molecule that plays critical roles in bacterial stress responses, biofilm formation, and virulence. A recent study identified gallein as a dual-specificity inhibitor against two families of PPK enzymes in Pseudomonas aeruginosa. This discovery may provide a new tool for further research on fundamental PPK and polyP functions in bacteria.
