The artemisinin analog SM934 alleviates dry eye disease in rodent models by regulating TLR4/NF-κB/NLRP3 signaling

Fang-ming Yang et al.

Summary: Research shows that SM934 has therapeutic potential for dry eye disease, as it increases tear secretion, maintains conjunctival goblet cells, reduces corneal damage, and decreases levels of inflammatory mediators. Additionally, SM934 reduces the accumulation of TLR4-expressing macrophages in the conjunctiva and suppresses the expression of inflammasome components.

Corneal neovascularization

Matthew P. Nicholas et al.

Summary: The optical clarity of the cornea is crucial for maintaining good visual acuity, while corneal neovascularization is a major cause of vision loss. Factors like infection, inflammation, hypoxia, trauma, corneal degeneration, and corneal transplantation can disrupt the homeostatic safeguards in the cornea that prevent angiogenesis.

Daphnetin inhibits spinal glial activation via Nrf2/HO-1/NF-rB signaling pathway and attenuates CFA-induced inflammatory pain

Yifan Yang et al.

Summary: Daphnetin shows significant alleviating effects on inflammatory pain induced by complete Freund's adjuvant in mice, through inhibition of spinal glial activation and reduced expression of spinal pro-inflammatory cytokines, possibly by negative regulation of the NF-rB pathway and activation of the Nrf2/HO-1 signaling pathway. These findings suggest the potential application of Daphnetin in treating chronic inflammatory pain.

Cucurbitacin B inhibits non-small cell lung cancer in vivo and in vitro by triggering TLR4/NLRP3/GSDMD-dependent pyroptosis

Renyikun Yuan et al.

Summary: The study revealed that CuB exerts anti-tumor effects in NSCLC by inducing pyroptosis through directly binding to Toll-like receptor 4 (TLR4) and activating the NLRP3 inflammasome, leading to Gasdermin D (GSDMD) cleavage. Additionally, CuB enhances mitochondrial reactive oxygen species (ROS) production, mitochondrial membrane protein Tom20 accumulation, and cytosolic calcium (Ca2+) release, ultimately promoting pyroptosis in NSCLC cells. A dosage-independent suppression of lung tumor growth was observed in mice treated with CuB, with 0.75 mg/kg showing a superior antitumor effect compared to Gefitinib.
