Pathophysiology of Hypertension The Mosaic Theory and Beyond

David G. Harrison et al.

Summary: Dr. Irvine Page proposed the Mosaic Theory of Hypertension in the 1940s, advocating that hypertension is the result of many interacting factors. Over the years, new concepts have arisen that allow further refinements of the theory, emphasizing the interdependence of various nodes.

Caveolar peroxynitrite formation impairs endothelial TRPV4 channels and elevates pulmonary arterial pressure in pulmonary hypertension

Zdravka Daneva et al.

Summary: Recent studies have found that the activity of TRPV4 channels in endothelial cell caveolae is impaired in patients with pulmonary hypertension and mouse models. Up-regulation of iNOS and NOX1 enzymes leads to the formation of the oxidant molecule peroxynitrite, which targets the structural protein caveolin-1 and reduces the activity of TRPV4 channels.

Selective cysteines oxidation in soluble guanylyl cyclase catalytic domain is involved in NO activation

Maryam Alapa et al.

Summary: The study found that redox modifications play a crucial role in NO activation of GC1, with NO-stimulated GC1 containing more bound Cys, potentially disulfide bonds. Mass spectrometry identified ten oxidized and two reduced Cys in NO-stimulated GC1, and computational modeling narrowed down potential Cys candidates involved in disulfide bonds to Cys489 and Cys571. Mutational studies confirmed the involvement of Cys489 and Cys571 in GC1's response to NO, potentially as a thiol/disulfide switch.
