Functional Comparison of Interferon-α Subtypes Reveals Potent Hepatitis B Virus Suppression by a Concerted Action of Interferon-α and Interferon-γ Signaling

Jieliang Chen et al.

Summary: The study identified IFN-alpha 14 as the most effective subtype in suppressing HBV replication, with significantly lower concentration values compared to conventional IFN-alpha 2. IFN-alpha 14 triggered a strong antiviral response through IFN-alpha and IFN-gamma signaling crosstalk, revealing mechanisms for potent HBV suppression. The findings deepen the understanding of the diverse activities of IFN-alpha subtypes and the synergistic effects of IFN-alpha and IFN-gamma signaling, potentially leading to improved HBV therapy strategies.

Rapid Turnover of Hepatitis B Virus Covalently Closed Circular DNA Indicated by Monitoring Emergence and Reversion of Signature-Mutation in Treated Chronic Hepatitis B Patients

Qi Huang et al.

Summary: The study retrospectively monitored the dynamics of lamivudine-resistant mutations in patients with HBV, finding a strong correlation between the genetic composition of cccDNA and serum HBV RNA. Results suggest that turnover of cccDNA in treated HBV patients occurs relatively rapidly, offering the potential for HBV cure through fully blocking cccDNA replenishment.

Analysis of Hepatitis B Virus Haplotype Diversity Detects Striking Sequence Conservation Across Genotypes and Chronic Disease Phase

Josef Wagner et al.

Summary: The study identified different haplotype populations associated with treatment outcomes and disease progression in chronic hepatitis B infection. Haplotype diversity varied across HBV genotypes and phases of CHB history. Highly conserved sequences in key genes were proposed as potential targets for antiviral therapies and predictors of treatment response.

Dicoumarol, an NQO1 inhibitor, blocks cccDNA transcription by promoting degradation of HBx

Sheng-Tao Cheng et al.

Summary: A novel small molecule targeting HBx was identified to combat chronic HBV infection, and it was revealed that NQO1 plays a role in HBV replication by regulating the stability of HBx protein.

Covalently closed circular DNA: The ultimate therapeutic target for curing HBV infections

Maria Guadalupe Martinez et al.

Summary: Current antiviral therapies for chronic hepatitis B can improve patients' quality of life but do not cure infected hepatocytes. Complete HBV cure requires therapies targeting the cccDNA pool directly. New technologies aim to achieve non-cytolytic direct cccDNA targeting for cure.
