Targeted Solvatochromic Fluorescent Probes for Imaging Lipid Order in Organelles under Oxidative and Mechanical Stress

Summary: A series of solvatochromic probes have been designed for specific targeting of different organelles in live cells, allowing the detection of variations in lipid order and quantitative description of lipid order in each organelle. These probes revealed the characteristics of organelles under stress conditions, with mitochondria and lysosomes particularly sensitive to stress. The new probes provide a powerful toolbox for monitoring cellular response to physical and chemical stimuli at the level of individual organelle membranes.

Synthesis and Transformations of NH-Sulfoximines

Summary: In recent years, there has been a significant increase in the use of sulfoximines in the field of chemistry, as they are considered valuable motifs in medicinal chemistry, due to pioneering works in clinical trials and the development of synthetic methods. Recent developments in sulfoximine synthesis, focusing on S-N and S-C bond formations since 2015, as well as the use of flow chemistry processes and subsequent transformations, particularly in N-functionalization, have been covered in this review.

Catalytic Enantioselective Synthesis of Pyridyl Sulfoximines

Photochemistry of N-Phenyl Dibenzothiophene Sulfoximine†

Summary: The study analyzed the photochemistry of N-phenyl dibenzothiophene sulfoximine, a popular scaffold in drug discovery, and found that it generates nitrenes and oxides under UV-A irradiation. The experiments demonstrated that in the presence of the nucleophilic agent DEA, N-phenyl dibenzothiophene sulfoximine produces azepines, while in the absence of DEA, it generates moderate amounts of azobenzene.

Late-stage functionalisation of alkyne-modified phospha-xanthene dyes: lysosomal imaging using an off-on-off type of pH probe

Summary: Near-infrared (NIR) fluorescent molecules are important for visualising biological processes, with P=O-substituted phospha-xanthene (POX) dyes being a promising scaffold due to their NIR emission and photostability. The development of POXs bearing a 4-ethynylphenyl group on the phosphorus atom allows for functionalisation with azide-tagged biomolecules via late-stage Cu-catalysed azide/alkyne cycloaddition (CuAAC), enabling target-selective labelling. A sophisticated NIR probe has been designed to have a bell-shaped pH-response and assess endosomal maturation, demonstrating the practical utility of functionalised POXs.

Visible light-promoted NH-halogenation of sulfoximines with dichloromethane or dibromomethane

Summary: In this study, dichloro- and dibromomethane were utilized as halogen sources to convert NH-sulfoximines to N-halo derivatives, using an in situ formed sulfoximidoyl-containing hypervalent iodine reagent under visible light. The reactions were carried out in air without the need for catalysts or additives, and the products were obtained in good to excellent yields.
