Inflammation in Atherosclerosis-No Longer a Theory

Peter Libby

Summary: Inflammation is closely linked to atherosclerosis, and recent clinical trials have shown the effectiveness of anti-inflammatory interventions in improving outcomes for patients with the disease. Further research is needed to optimize these interventions and improve patient selection.

The changing landscape of atherosclerosis

Peter Libby

Summary: Emerging evidence has led to a significant evolution of concepts related to atherosclerosis, with a shifting risk profile that is no longer limited to specific regions or demographics. Recent research has highlighted new risk factors and mechanisms underlying atherosclerosis, opening doors to potential therapeutic interventions for this widespread disease.

Plaque erosion and acute coronary syndromes: phenotype, molecular characteristics and future directions

Akl C. Fahed et al.

Summary: Atherosclerotic plaque erosion is increasingly recognized as a common feature in acute coronary syndromes. Recent research suggests that conservative treatment may be an option for patients with plaque erosion. Advanced imaging techniques and basic science experiments have provided insights into the characteristics of plaque erosion.

Targeting Inflammatory Pathways in Cardiovascular Disease: The Inflammasome, Interleukin-1, Interleukin-6 and Beyond

Peter Libby

Summary: Inflammation plays a causal role in human atherosclerosis, and recent clinical trials point towards potential novel treatments. This review evaluates the current state of anti-inflammatory interventions, focusing on innate immunity and utilizing biomarkers to guide therapy allocation. The validation of decades of research in recent clinical investigations provides a strong motivation for further efforts to target inflammation in atherosclerosis.

Cell-specific and divergent roles of the CD40L-CD40 axis in atherosclerotic vascular disease

Michael Lacy et al.

Summary: The CD40L-CD40 signaling axis plays a role in atherosclerosis. This study investigates the cell-specific functions of the most relevant CD40L-expressing cell types. Deficiency of T cell-derived CD40L reduces and stabilizes plaques through impaired Th1 polarization, while platelet-derived CD40L ameliorates atherothrombosis.

The Role of SGLT2 Inhibitors in Vascular Aging

Le Liu et al.

Summary: Vascular aging is characterized by organic and functional changes in blood vessels, involving factors such as decline in autophagy levels, DNA damage, MicroRNA, oxidative stress, and impacts arterial morphology and cellular level. SGLT2 inhibitors can delay vascular aging, improve vascular function, and provide theoretical support and guidance for related diseases.

GLP-1 receptor agonists (GLP-1RAs): cardiovascular actions and therapeutic potential

Xiaoxuan Ma et al.

Summary: GLP-1RAs are a type of anti-diabetic medication that stimulate insulin secretion, lower blood glucose levels, and have positive effects on cardiovascular protection without causing side effects such as weight gain and hypoglycemia. Numerous cardiovascular outcomes trials have supported the overall cardiovascular benefits of GLP-1RAs, which also lower plasma lipid levels and blood pressure, contributing to a reduction in atherosclerosis and cardiovascular diseases.
