StrVCTVRE: A supervised learning method to predict the pathogenicity of human genome structural variants

Andrew G. Sharo et al.

Summary: This article introduces a method called StrVCTVRE to distinguish between pathogenic and benign structural variants (SVs). By integrating multiple features and using a rare training set for classification, this method reduces about half of the SVs while maintaining a high sensitivity. It provides support for further investigation into unresolved cases and understanding new mechanisms of disease.

Haplotype-resolved diverse human genomes and integrated analysis of structural variation

Peter Ebert et al.

Summary: Through the use of long-read and strand-specific sequencing technologies, this study has achieved the de novo assembly of high-quality haplotype-resolved human genomes without the need for parent-child trio data. The research identified various forms of genetic variation, including structural variants and SV hotspots, and revealed the mechanisms of SV formation while providing SV candidates for adaptive selection within the human population.

Heterozygous ANKRD17 loss-of-function variants cause a syndrome with intellectual disability, speech delay, and dysmorphism

Maya Chopra et al.

Summary: Loss of ANKRD17 leads to a neurodevelopmental disorder characterized by developmental delay, intellectual disability, growth failure, speech impairment, along with other features like feeding difficulties, epilepsy, ophthalmological abnormalities, and joint hypermobility. Analysis suggests that damaging ANKRD17 variants disrupt the stability of the protein and contribute to the observed phenotypes.
