Alpha-Tocopherol Metabolites (The Vitamin E Metabolome) and Their Interindividual Variability during Supplementation

Desiree Bartolini et al.

Summary: New metabolomics protocols have allowed for the exploration of individual differences in the metabolism of alpha-tocopherol, revealing marked variability in the metabolites. Enzymatic metabolites showed significant interindividual variability, while free radical-derived metabolite alpha-tocopheryl quinone demonstrated a significant correlation with post-supplementation levels of alpha-tocopherol. The study also found that the expression of certain target genes of alpha-tocopherol in leukocytes was stimulated by supplementation, highlighting the importance of considering intrinsic variability in future studies.

Factors affecting in vitro and in vivo antioxidant effects: Experimental conditions and nature of oxidants determine antioxidant efficacy

Etsuo Niki

Summary: The role of antioxidants in health maintenance and disease prevention has received significant attention due to the implications of reactive oxygen and nitrogen species in various diseases. The effectiveness of antioxidants is influenced by multiple factors under different reaction conditions and media. Researchers focus on experimental conditions and interpretation of results to understand the beneficial effects and limitations of antioxidants against detrimental oxidation of biological molecules, with a particular emphasis on cell culture experiments and the diverse effects of multiple oxidants on antioxidant efficacy.

Deep mining of oxysterols and cholestenoic acids in human plasma and cerebrospinal fluid: Quantification using isotope dilution mass spectrometry

Eylan Yutuc et al.

Summary: The method described allows for identification and quantification of multiple sterols in plasma and cerebrospinal fluid, utilizing isotope-dilution mass spectrometry for absolute quantification. It is applicable for semi-quantification of other sterols without isotope labelled surrogates and approximate quantification of partially identified sterols. The method demonstrated its value in confirming sterol phenotypes in patients with specific disorders.

Lipid peroxidation products as a mediator of toxicity and adaptive response - The regulatory role of selenoprotein and vitamin E

Yoshiro Saito

Summary: Lipid peroxidation and its products play a significant role in physiological and pathophysiological conditions, with mechanisms including enzymatic oxidation, free radical-mediated oxidation, and nonradical-mediated oxidation. The products of lipid peroxidation can lead to cytotoxicity and novel biological activity, while the antioxidant system, particularly selenoproteins and vitamin E, plays a crucial role in preventing lipid peroxidation and ferroptosis.

Plasma lipidome profiling of newborns with antenatal exposure to Zika virus

Nieli Rodrigues da Costa Faria et al.

Summary: The study compared the plasma lipidome profiles of newborns with microcephaly and normocephalic newborns exposed to ZIKV with non-infected controls. The results suggest that ZIKV exposure leads to alterations in plasma lipidome, which could potentially serve as indicators for identification and monitoring of antenatal ZIKV exposure.

Lipid peroxidation as measured by chromatographic determination of malondialdehyde. Human plasma reference values in health and disease

Summary: Oxidative stress, caused by an imbalance between prooxidants and antioxidants, can lead to biomolecular damage and development of diseases. Lipids are sensitive to oxidant attack, with MDA, 4-HNE, and F2-isoprostane being the main biomarkers for lipid peroxidation assessment. The profile of PUFAs in tissues determines the proportion of each biomarker.
