Geometric Lessons and Design Strategies for Nanoscale Protein Cages

Joshua Laniado et al.

Summary: Protein molecules add rich functionality to nanoscale architectures, particularly in high-symmetry protein cages. This study introduces a method for designing protein assemblies, successfully creating cubically symmetric cages through a combination of two simpler symmetries. Experimental testing revealed the formation of the intended octahedral cage, while another design resulted in a collapsed structure of lower stoichiometry and symmetry, providing insights for improved creation of nanoscale protein architectures.

Toward complete rational control over protein structure and function through computational design

Jared Adolf-Bryfogle et al.

Summary: The grand challenge of protein design lies in producing polypeptides with arbitrary functionality, conformation, and biochemical properties through a general method. Various methods have been developed to improve native proteins, design ideal proteins de novo, and redesign suboptimal proteins. While there are still unsolved problems in manipulating protein-protein interfaces and molecular catalysis, promising results indicate that a general approach to manipulating polypeptides is within reach.

Nanopore-Controlled Dual-Surface Modifications on Artificial Protein Nanocages as Nanocarriers

Erika Nasu et al.

Summary: Chemical modification of protein nanocages holds promise for various applications, but dual-surface modification using different chemicals remains challenging. Surface pores on artificial protein nanocage TIP60 function as size-dependent molecular filters, allowing only small molecules to pass through. Dual-surface functionalization of TIP60 enables it to release internal small molecules in a redox-responsive manner, making it a useful nanocarrier for drug delivery and molecular filtration systems.
