COVID-19: Discovery, diagnostics and drug development

Tarik Asselah et al.

Summary: COVID-19 started in Wuhan in 2019 and has become a global pandemic. The virus responsible, SARS-CoV-2, shares a high sequence identity with bat- and pangolin-derived coronaviruses. Risk factors for SARS-CoV-2 infection include age, obesity, diabetes, and hypertension.

ApoE-Isoform-Dependent SARS-CoV-2 Neurotropism and Cellular Response

Cheng Wang et al.

Summary: The study suggests a potential causal role of ApoE4 in the severity of COVID-19, as tested in human-induced pluripotent stem cell (hiPSC) models for SARS-CoV-2 neurotropism. Increased infection rates of SARS-CoV-2 in ApoE4/4 neurons and astrocytes were observed, with changes in ApoE4 astrocytes upon infection. Remdesivir treatment was effective in inhibiting SARS-CoV2 infection in hiPSC neurons and astrocytes, highlighting the impact of risk factors on COVID-19 severity.

Summary: This study examined the longitudinal brain metabolic pattern in COVID-19-related encephalopathy using 18F-FDG-PET/CT. It found a consistent pattern of hypometabolism in various brain regions, with patients presenting with cognitive and behavioral disorders in the acute phase and ongoing cognitive and emotional symptoms 6 months after disease onset.

Early transmissibility assessment of the N501Y mutant strains of SARS-CoV-2 in the United Kingdom, October to November 2020

Kathy Leung et al.

Summary: Two new lineages of SARS-CoV-2 with the N501Y mutation in the receptor-binding domain of the spike protein have spread rapidly in the United Kingdom. The 501Y lineage without amino acid deletion Delta 69/Delta 70 was estimated to be 10% more transmissible than the 501N lineage, while the 501Y lineage with amino acid deletion Delta 69/Delta 70 was estimated to be 75% more transmissible than the 501N lineage.

Summary: A retrospective study examining patients with encephalopathy associated with COVID-19 found that laboratory markers of COVID-19 severity were common, but did not correlate with the severity of encephalopathy. Neuroimaging and CSF analysis did not support direct viral CNS invasion or CNS inflammation as the cause of encephalopathy.

The role of Neuropilin-1 in COVID-19

Bindu S. Mayi et al.

Summary: NRP-1, a member of a family of signaling proteins, plays a critical role in SARS-CoV-2 infection and may be involved in immune function, transmission pathways, and disease severity. Research suggests NRP-1 could serve as a therapeutic target in SARS-CoV-2 infections, highlighting the need for further study.

Susceptibility-weighted imaging reveals cerebral microvascular injury in severe COVID-19

John Conklin et al.

Summary: In patients with severe COVID-19, microvascular brain lesions, characterized by punctate SWI lesions, are commonly found in the subcortical and deep white matter. These lesions are associated with widespread microvascular injury, indicating a risk for hemorrhagic and ischemic lesions in the brain.

Neuroimaging the Neuropathogenesis of HIV

Anna H. Boerwinkle et al.

Summary: Recent neuroimaging studies have provided insight into the structural, functional, and molecular changes occurring in the brain due to HIV. The findings suggest that HIV triggers a strong neuroimmune response and may lead to a cascade of events including increased chronic inflammation and cognitive decline. These outcomes are further exacerbated by age and age-related comorbidities, as well as lifestyle factors such as drug use/abuse.

Summary: Research findings suggest that pericytes may serve as important portals for SARS-CoV-2 infection in the nervous system. Introducing pericyte-like cells (PLCs) into cortical organoids can provide an experimental model that supports virus entry and replication.
