STIM-Orai1 signaling regulates fluidity of cytoplasm during membrane blebbing

Kana Aoki et al.

Summary: The cytoplasm in mammalian cells is considered homogeneous. Here authors report that the cytoplasmic fluidity is regulated in the blebbing cells, which is regulated by calcium concentration in the expanding blebs and involves the STIM-Orai1 pathway.

Mechanisms of nonvesicular lipid transport

Karin M. Reinisch et al.

Summary: Lipids can be transported between cellular membranes via nonvesicular mechanisms, supporting biogenesis of rapidly expanding membranes or allowing targeted modulation of specific lipid levels. Nonvesicular transport likely occurs at membrane contact sites and involves lipid transport proteins, but many questions remain unanswered regarding the mechanisms involved.

Lymphocyte egress signal sphingosine-1-phosphate promotes ERM-guided, bleb-based migration

Tanner F. Robertson et al.

Summary: This study elucidates the role of ERM proteins in regulating S1P-dependent T cell egress, showing that ERM-deficient T cells exhibit impaired migration induced by S1P but not by standard protein chemokines. The distinct modes of motility induced by S1P and chemokines are specialized for T cell migration across lymphatic barriers and through tissue stroma, respectively.

Nuclear fragility, blaming the blebs

Nishit Srivastava et al.

Summary: The cell nucleus can adopt a variety of shapes in tissues due to cell deformities caused by cell crowding or migration through dense matrices. Recent studies have shown that the nuclear envelope can form blebs, leading to nuclear envelope opening and uncontrolled nucleocytoplasmic mixing. While blebs are a major source of nuclear instability, they are poorly understood and require more in-depth research.

Molecular basis of functional exchangeability between ezrin and other actin-membrane associated proteins during cytokinesis

Guang Yang et al.

Summary: The study reveals the molecular basis of exchangeability between ezrin and other actin-membrane associated proteins during cytokinesis, showing specific designs of actin- or membrane-associated moieties with limited exchangeability. Depletion of anillin and supervillin leads to the overaccumulation of different domains of ezrin at the cleavage furrow, suggesting differential binding sites of ezrin with these proteins. Chimeric mutants demonstrate that ezrin's C-terminal domain can substitute for anillin domains in cytokinesis and cell proliferation, while anillin domains cannot substitute for ezrin domains in controlling cortical activities.

ADF and cofilin-1 collaborate to promote cortical actin flow and the leader bleb-based migration of confined cells

Maria F. Ullo et al.

Summary: The study reveals that Cofilin-1 and ADF are essential actin remodeling factors for fast amoeboid migration in melanoma cells. Cofilin-1 regulates cortical actin by enriching at the necks of leader blebs, while ADF affects bleb sizes and cell motility frequency.

Crosstalk between mechanotransduction and metabolism

Patrizia Romani et al.

Summary: Mechanical forces play a crucial role in regulating cell function and behavior by modulating cell metabolism and impacting tissue mechanics. Recent evidence suggests that mechanical cues from the extracellular matrix and cell adhesion complexes influence metabolic pathways, highlighting the reciprocal crosstalk between cell mechanics and metabolism.
