Preparation and in vitro characterization of histidine trimethyl chitosan conjugated nanocomplex incorporated into injectable thermosensitive hydrogels for localized gene delivery

Summary: In this study, localized and sustained delivery of DNA using biomaterial scaffolds was achieved by synthesizing HTMC and fabricating nanocomplexes with pDNA, which were then loaded into hydrogel scaffolds. Transfection experiments demonstrated that HTMC/pDNA complexes remained stable and active inside the hydrogel, although the transfection efficacy slightly decreased after incorporation into the hydrogel.
