SOD1-interacting proteins: Roles of aggregation cores and protein degradation systems

Summary: Cu/Zn superoxide dismutase (SOD1) mutations are associated with amyotrophic lateral sclerosis (ALS). Using a proteomics approach, the study compared protein expression in spinal cord homogenates from non-transgenic (NTG) and ALS model mice to identify proteins interacting with SOD1 seeds. Various SOD1-interacting proteins involved in protein quality control, cytoskeletal organization, growth factor signaling pathways and downstream cascades were identified, indicating complex interactions between pathways that may lead to fatal cellular dysfunction in ALS.

The method enabled highly selective quantification, enhanced sequence coverage and structural characterization of endogenous TDP-43, providing a valuable tool for research on frontotemporal dementia and amyotrophic lateral sclerosis.

Defining the Caprin-1 Interactome in Unstressed and Stressed Conditions

Summary: This study identified 1543 proteins interacting with Caprin-1, primarily associated with ribosome, spliceosome, and RNA transport pathways under stressful conditions. Four Caprin-1 interactors were validated in arsenite-induced stress granules, and SNRNP200 was found in osmotic- and thermal-induced stress granules as well. Additionally, SNRNP200 was identified in cytoplasmic aggregates in amyotrophic lateral sclerosis (ALS) patient neurons.

Network Analysis of the CSF Proteome Characterizes Convergent Pathways of Cellular Dysfunction in ALS

Summary: This study compared CSF proteomic data from ALS patients, Parkinson's disease patients, and healthy controls, revealing alterations in immune system and axonal modules in ALS, which are associated with TDP-43 dysfunction. Longitudinal analysis showed temporal changes in the CSF proteome in ALS patients. Protein correlation network disruptions in Parkinson's disease did not show any module enrichment.
