APR-246 induces early cell death by ferroptosis in acute myeloid leukemia

Rudy Birsen et al.

Summary: APR 246 is a promising therapeutic agent for myelodysplastic syndromes and acute myeloid leukemia that targets p53 mutated proteins. It has been found to reactivate the transcriptional activity of p53 mutants and induce p53-independent cell death. The study also demonstrates the association between APR 246 and ferroptosis, a recently described cell death process. The detoxification capacity of AML cells to maintain glutathione biosynthesis may play a key role in determining sensitivity to APR 246.

Resolving the paradox of ferroptotic cell death: Ferrostatin-1 binds to 15LOX/PEBP1 complex, suppresses generation of peroxidized ETE-PE, and protects against ferroptosis

Tamil S. Anthonymuthu et al.

Summary: HpETE-PE is a cell death signal in ferroptosis, produced by the 15LOX/PEBP1 complex. Fer-1 does not directly affect 15LOX, but can effectively inhibit the production of HpETE-PE by the 15LOX/PEBP1 complex. This study resolves the long-standing Fer-1 anti-ferroptotic paradox.

Ferroptosis: molecular mechanisms and health implications

Daolin Tang et al.

Summary: Cell death can be executed through different subroutines, such as ferroptosis which is an iron-dependent form of non-apoptotic cell death. Ferroptosis can occur through two major pathways, driven by redox imbalance and abnormal expression of redox-active enzymes. The process is precisely regulated at multiple levels, with the transcription factor NFE2L2 playing a central role in anti-ferroptotic defense.

Membrane Damage during Ferroptosis Is Caused by Oxidation of Phospholipids Catalyzed by the Oxidoreductases POR and CYB5R1

Bo Yan et al.

Summary: Ferroptosis is a form of necrotic cell death caused by iron-dependent peroxidation of polyunsaturated phospholipids on cell membranes, which is carried out by oxidoreductases transferring electrons to oxygen to generate hydrogen peroxide for lipid peroxidation. This process needs to be counteracted by antioxidant systems to prevent cell death.

Acid sphingomyelinase-dependent autophagic degradation of GPX4 is critical for the execution of ferroptosis

Faisal Thayyullathil et al.

Summary: This study reveals a novel role of ASM in promoting ferroptosis through activation and ceramide generation, which are necessary for ROS and LPO induced by Era. NADPH oxidase-derived ROS regulates ASM-initiated redox signaling in a positive feedback manner, while ASM-mediated activation of autophagy plays a crucial role in GPX4 degradation and subsequent ferroptosis. Moreover, genetic activation of ASM increases ferroptosis in cancer cells induced by various FINs, suggesting ASM could be a potential target for improving pathological conditions related to ferroptosis.

The Lipogenic Regulator SREBP2 Induces Transferrin in Circulating Melanoma Cells and Suppresses Ferroptosis

Xin Hong et al.

Summary: Circulating tumor cells shed by cancer into the bloodstream coordinately upregulate lipogenesis and iron homeostasis pathways, contributing to resistance to BRAF inhibitors. The lipogenesis regulator SREBP2 induces transcription of iron carrier TF, reducing oxidative stress and conferring resistance to ferroptosis inducers. Presence of CTCs with high lipogenic and iron metabolic signatures is correlated with adverse clinical outcome in melanoma patients.

Knockdown of DAZAP1 inhibits proliferation, migration, and invasion of HCC cells, and also reduces cellular sensitivity to SF. Furthermore, DAZAP1 is identified as an inhibitor of ferroptosis and positively regulates the stability of SLC7A11 mRNA, potentially offering new insights into biomarkers and therapeutic targets in HCC patients.

HO-1-mediated ferroptosis as a target for protection against retinal pigment epithelium degeneration

Zhimin Tang et al.

Summary: Research has identified that oxidative stress-induced RPE degeneration is mainly regulated by HO-1-mediated ferroptosis, controlled by the Nrf2-SLC7A11-HO-1 pathway. In mouse experiments, inhibiting HO-1 can block RPE ferroptosis, and using the HO-1 inhibitor ZnPP effectively rescues RPE degeneration with superior therapeutic effects. This highlights that targeting HO-1-mediated RPE ferroptosis could serve as an effective retinal-protective strategy for preventing retinal degenerative diseases, including AMD.

Targeted reduction of cholesterol uptake in cholesterol-addicted lymphoma cells blocks turnover of oxidized lipids to cause ferroptosis

Jonathan S. Rink et al.

Summary: Recent research suggests that malignant cells dependent on cholesterol uptake from lipoproteins are susceptible to ferroptosis, a type of cell death triggered by accumulation of oxidized lipids in cell membranes. Treatment of these cells with HDL NPs activates a compensatory metabolic response, leading to increased de novo cholesterol synthesis and downregulation of GPX4 expression, resulting in ferroptosis-mediated cell death. In vivo studies in mouse lymphoma xenografts and patient samples further support the potential of targeting SCARB1 with HDL NPs for cholesterol uptake-addicted lymphoma cells.
