SP1-independent inhibition of FOXM1 by modified thiazolidinediones

Summary: By modifying the thiazolidinedione scaffold, researchers developed drugs capable of binding to and inhibiting the in vitro transcriptional activity of the oncogenic protein FOXM1. The most active molecule identified, compound TFI-10, showed potential binding interactions with specific amino acids and does not target the in vitro expression of Sp1, suggesting the possibility of designing FOXM1 inhibitors with improved selectivity.

Summary: The study revealed that FOXM1 was highly upregulated in psoriatic skin tissues and TNF-alpha-stimulated HaCaT cells. Knockdown of FOXM1 inhibited cell proliferation and promoted apoptosis in HaCaT cells, while also suppressing the release of inflammatory cytokines. FOXM1 mediated its effects through the NF-kappa B pathway, regulating cell proliferation, apoptosis, and inflammation in response to TNF-alpha.

Recurrence-free interval 12 months after local treatment of mast cell tumors in dogs using intratumoral injection of tigilanol tiglate

Through high-throughput screening of a library of around 4000 compounds, researchers identified compounds that can activate human macrophages towards specific polarization states, with some capable of reprogramming macrophages between different states. Transcriptional analysis helped uncover the pathways and targets through which these compounds modulate macrophage activation.

Thiostrepton inhibits growth and induces apoptosis by targeting FoxM1/SKP2/MTH1 axis in B-precursor acute lymphoblastic leukemia cells

Summary: The study showed that downregulation of FoxM1 by its inhibitor thiostrepton inhibited cell viability and induced caspase-dependent apoptosis in B-pre-ALL cell lines. Thiostrepton led to downregulation of FoxM1 and multiple cancer-related genes, increased Bax/Bcl2 ratio, and suppression of antiapoptotic proteins. Co-treatment of B-pre-ALL with subtoxic doses of thiostrepton and bortezomib potentiated proapoptotic action, suggesting that targeting FoxM1 expression could be an attractive strategy for B-pre-ALL treatment.

Newly established patient-derived organoid model of intracranial meningioma

Summary: This study established 18 organoid models of meningiomas and identified upregulated FOXM1 as a potential treatment target for meningioma. Overexpression of FOXM1 in benign meningiomas increased proliferation, while depletion of FOXM1 in malignant meningiomas decreased proliferation. FOXM1 inhibitor thiostrepton combined with radiation therapy significantly inhibited the proliferation of malignant meningioma organoid models.

Liposomal Thiostrepton Formulation and Its Effect on Breast Cancer Growth Inhibition

Summary: Liposome-encapsulated thiostrepton (TSLP) showed a higher potential in reducing FOXM1 levels in MCF-7 breast cancer cells compared to free thiostrepton. TSLP improved the efficiency and specificity of thiostrepton in reducing cell viability of MCF-7 cells, without affecting fibroblast cells. TSLP also had the ability to induce cell death in both 2D monolayer and 3D spheroid cultures of MCF-7 cells.

Dual inhibition of FOXM1 and its compensatory signaling pathway decreased the survival of ovarian cancer cells

Summary: The study aimed to analyze compensatory signaling pathways induced by FDI-6, a FOXM1 inhibitor, in ovarian cancer cells and evaluated the effectiveness of simultaneous inhibition of FOXM1 and the compensatory signaling pathway in decreasing cancer cell survival. The study identified proteins involved in the compensatory mechanism activated by FDI-6 and found that combining FDI-6 with inhibitors of N-Ras or p-PKC delta (S664) significantly improved the reduction in ovarian cancer cell viability compared to FDI-6 alone.
