Alternative splicing modulates cancer aggressiveness: role in EMT/metastasis and chemoresistance

Debanwita Roy Burman et al.

Summary: Enhanced metastasis and disease recurrence contribute to the high mortality rates in cancer. Epithelial-Mesenchymal Transition (EMT) and alternative splicing play crucial roles in cancer invasiveness, drug resistance, and stem-like behavior. Splicing factors control the splicing alterations of genes like FGFR and CD44 during trans-differentiation, impacting cancer progression.

Arsenic-induced epigenetic changes in cancer development

Wesley N. Saintilnord et al.

Summary: Arsenic, a ubiquitous metalloid, poses major health concerns worldwide due to its high toxicity, potentially driving oncogenic gene expression through mechanisms such as generation of reactive oxygen species or epigenetic dysregulation. Further research is needed to fully understand the role of arsenic in cancer development.

Alternative splicing acts as an independent prognosticator in ovarian carcinoma

Yan Ouyang et al.

Summary: This study identified survival-associated alternative splicing events in ovarian cancer, constructed an independent prognostic signature, and revealed potential regulatory mechanisms between splicing factors and candidate AS events. The prognostic signature based on seven types of AS events demonstrated powerful efficiency in predicting patient outcomes. Additionally, survival-related AS events were found to be involved in tumor-related pathways and certain splicing factors were correlated with prognosis-related AS events.

Dynamic alteration in miRNA and mRNA expression profiles at different stages of chronic arsenic exposure-induced carcinogenesis in a human cell culture model of skin cancer

Mayukh Banerjee et al.

Summary: Chronic arsenic exposure can lead to skin cancer, with altered microRNA and mRNA expression playing a key role in carcinogenesis. Using a HaCaT cell line model, changes in genome-wide differential expression of miRNA and mRNA were investigated at 3 strategic time points upon exposure to chronic sodium arsenite. Pathway analyses revealed dysregulation of carcinogenesis-related pathways and networks in a complex coordinated manner at each time point.

Multilayered regulation of proteome stoichiometry

Koji Ishikawa

Summary: Cellular systems rely on specific stoichiometries of subunit proteins in multiprotein complexes. To maintain proper stoichiometry, protein synthesis and degradation are controlled even in the presence of genetic perturbations caused by changes in gene dosage. Excess unassembled subunits synthesized due to increased gene copy number are rapidly degraded by the ubiquitin-proteasome system, a mechanism known as protein-level dosage compensation.

RNA in cancer

Gregory J. Goodall et al.

Summary: Recent findings have shown that RNA processing is systematically altered in cancer, with mutations in RNA splicing factor genes and the shortening of 3' untranslated regions widely observed. Additionally, evidence suggests that other types of RNAs, including circular RNAs, can contribute to tumorigenesis.
