Summary: Reports on the impacts of per- and polyfluoroalkyl substances (PFAS) on the environment and human health have increased substantially in peer-reviewed literature. Epidemiological studies have shown associations between exposure to specific PFAS and various health effects, and further research is needed to understand differences in responses between sexes, species, and life stages. More contemporary approaches such as read-across, molecular dynamics, and protein modeling are proposed to accelerate the development of toxicity information on PFAS, as well as the need for precaution to protect human health given the known effects of certain PFAS compounds.

Recent US State and Federal Drinking Water Guidelines for Per- and Polyfluoroalkyl Substances

Summary: PFAS are a class of synthetic chemicals that have been produced for over 70 years, with increasing concerns due to their widespread environmental presence, extreme persistence, bioaccumulative nature, and evidence for health effects from environmentally relevant exposures. Multiple US states have found the USEPA Health Advisories insufficiently protective and have developed more stringent guidelines for PFOA, PFOS, and other PFAS, with additional state guidelines expected to be released. The variations in state guidelines for PFAS reflect differences in toxicity and exposure considerations, with the range not unexpected given the different scientific approaches and historical context.

Summary: HFPO-DA is a developmental toxicant in rats, causing biological changes in maternal and fetal livers, decreased pup birth weight and increased neonatal mortality, as well as metabolic abnormalities at birth. The potency of HFPO-DA in rats is comparable to PFOS based on serum concentration, and the spectrum of adverse effects is consistent with other PFAS such as PFOS and PFOA.

Diet is a major pathway of human exposure to some PFAS, but there is significant variability across populations and compounds. Total fluorine data and information on unidentified organofluorine are essential, and water has been identified as a major exposure source in contaminated communities. As water sources are remediated, other sources of exposure such as dust, personal care products, and indoor environments may become more significant.
