Simvastatin modulates estrogen signaling in uterine leiomyoma via regulating receptor palmitoylation, trafficking and degradation

Sadia Afrin et al.

Summary: The study found that simvastatin can significantly reduce E-2-induced proliferation and PCNA expression, as well as decrease total ER-alpha expression in leiomyoma cells. Additionally, simvastatin alters the subcellular localization of ER-alpha by inhibiting its trafficking, and inhibits E-2 downstream signaling, ER/ER activity, and responsive genes.

Wnt/β-catenin signaling pathway in uterine leiomyoma: role in tumor biology and targeting opportunities

Malak El Sabeh et al.

Summary: Uterine leiomyoma is the most common tumor of the female reproductive system, with poorly understood underlying mechanisms, but alterations in signaling pathways are key players in its pathobiology. The Wnt/beta-catenin pathway plays a crucial role in leiomyoma development, with specific molecules within this pathway showing potential as therapeutic targets.

Uterine Stem Cells and Benign Gynecological Disorders: Role in Pathobiology and Therapeutic Implications

Malak El Sabeh et al.

Summary: This review summarizes the involvement of stem cells in the pathogenesis of benign gynecological conditions, focusing on their role in leiomyomas, endometriosis, and adenomyosis. The conversion of uterine stem cells to tumor initiating stem cells in these conditions, as well as their clinical implications, are discussed. Despite the significant burden these diseases pose, the understanding of the role of stem cells in their development is still incomplete.
