The H+-ATPase (V-ATPase): from proton pump to signaling complex in health and disease

Amity F. Eaton et al.

Summary: The primary function of H+-ATPase is to create an electrochemical proton gradient across eukaryotic cell membranes to energize cellular processes. It acidifies intracellular vesicles and organelles for essential cell biological events. Additionally, V-ATPases play central roles in normal and pathophysiological processes that impact human health, influencing areas such as cancer, neurodegenerative diseases, and sensory perception.

The Ncoa7 locus regulates V-ATPase formation and function, neurodevelopment and behaviour

Enrico Castroflorio et al.

Summary: Members of the Tre2/Bub2/Cdc16 (TBC), lysin motif (LysM), domain catalytic (TLDc) protein family are associated with neurodevelopmental disorders, including NCOA7 linked to autism. NCOA7 is shown to regulate the function of the vacuolar (V)-ATPase in the brain, with its deletion resulting in neuronal developmental defects and lysosomal dysfunction. Behavioral assessment of Ncoa7 deletion mice revealed anxiety and social defects, indicating the importance of NCOA7 in neurodevelopment.

Crystal structure of the TLDc domain of human NCOA7-AS

Mary Arnaud-Arnould et al.

Summary: The TLDc domain is associated with oxidation resistance related functions and well conserved among eukaryotes. Proteins with a TLDc domain in humans, such as NCOA7 and NCOA7-AS, play a protective role against oxidative stress and interact with subunits of the vacuolar proton pump V-ATPase to impair viral infection through the endosomal pathway. The crystal structure of the TLDc domain from NCOA7 and NCOA7-AS has been reported at 1.8 A resolution, providing insights into its function and potential therapeutic applications.
