Milk exosomes: A biogenic nanocarrier for small molecules and macromolecules to combat cancer

Raghuram Kandimalla et al.

Summary: Exosomes are biogenic nanocarriers of endocytic origin found in biofluids like milk, with milk exosomes being a particularly promising source for drug delivery. They are being increasingly explored for delivering both small drug molecules and siRNA to combat cancer, thanks to their biocompatibility and unique properties. Various isolation techniques, physicochemical properties, biodistribution, and applications of milk exosomes in drug delivery were highlighted in this article.

Bovine Milk-Derived Exosomes as a Drug Delivery Vehicle for miRNA-Based Therapy

Lorena del Pozo-Acebo et al.

Summary: miRNAs are potential mediators of gene expression in therapeutic strategies, but the instability of extracellular RNAs limits their biological function. Strategies to enhance miRNA therapeutic potential include developing efficient, tissue-specific delivery methods using exosomes. However, a standard, reproducible, and cost-effective protocol for exosome isolation and extracellular RNA delivery is currently lacking.

Microbubbles versus Extracellular Vesicles as Therapeutic Cargo for Targeting Drug Delivery

Mujib Ullah et al.

Summary: Extracellular vesicles (EVs) and microbubbles are both nanoparticles in drug-delivery systems with potential as drug carriers for therapeutic targets. They have shown capability in delivering chemotherapeutic drugs to tumor and damaged tissues. However, there is a lack of standards for evaluating the benefits of EVs (natural carrier) versus microbubbles (synthetic carrier), as well as limited research on their targeted delivery or efficacy. In this Perspective, we compare EVs and microbubbles to understand their utility in delivering drugs to their site of action and future clinical translation.

Endothelial-Derived miR-17∼92 Promotes Angiogenesis to Protect against Renal Ischemia-Reperfusion Injury

Takuto Chiba et al.

Summary: miR-17 similar to miR-92 is dynamically regulated in renal endothelial cells after renal IRI and plays a role in the pathogenesis of AKI. Endothelial-specific miR-17 similar to miR-92 knockout exacerbates renal IRI, promoting renal tubular injury, reducing renal blood flow, and increasing oxidative stress in the kidneys. miR-17 similar to miR-92 is critical in the angiogenic response after renal IRI, and treatment with miR-18a and miR-19b mimics can mitigate the damage.

M2 Macrophage-Derived Exosomes Promote Angiogenesis and Growth of Pancreatic Ductal Adenocarcinoma by Targeting E2F2

Yuhan Yang et al.

Summary: The study revealed that exosomes derived from M2 macrophages can promote angiogenesis in PDAC, leading to accelerated tumor growth. By transferring miR-155-5p and miR-221-5p to endothelial cells, the interaction between TAMs and angiogenesis in PDAC was further confirmed. Targeting exosomal miRNAs from TAMs may provide diagnostic and therapeutic strategies for PDAC.

Targeted Oral Delivery of Paclitaxel Using Colostrum-Derived Exosomes

Raghuram Kandimalla et al.

Summary: Paclitaxel (PAC) is widely used in the treatment of various cancers, including lung cancer, but its solvent-based formulation is associated with severe side effects. Utilizing bovine colostrum-derived exosomes as a delivery system for PAC can enhance therapeutic efficacy and reduce toxicity, providing a promising alternative to current therapies. Studies show that a tumor-targeted oral formulation of PAC delivered by exosomes significantly improves efficacy and safety compared to traditional intravenous administration.
