Cystine-glutamate antiporterxCTas a therapeutic target for cancer

Lin Liu et al.

Summary: xCT, a component of the cystine/glutamic acid reverse transporter, protects cells from oxidative stress and ferroptosis by promoting cystine uptake and glutathione biosynthesis. Its high expression in various tumors makes it a potential target for cancer treatment.

Centipedegrass extract enhances radiosensitivity in melanoma cells by inducing G2/M cell cycle phase arrest

Seong Hee Kang et al.

Summary: Melanoma is an aggressive and potentially fatal cancer with low sensitivity to chemotherapy and radiation therapy. The study found that centipedegrass extract combined with ionizing radiation increased apoptotic cell death and cell cycle arrest in melanoma cells, enhancing radiosensitivity via the MAPKs pathway.

Long noncoding RNA LINC00518 induces radioresistance by regulating glycolysis through an miR-33a-3p/HIF-1α negative feedback loop in melanoma

Yan Liu et al.

Summary: LINC00518 is a long noncoding RNA that is highly expressed in various cancers, including cutaneous malignant melanoma (CMM), and plays a role in promoting cancer progression. In CMM, LINC00518 induces radioresistance by regulating the glycolysis pathway through a negative feedback loop involving miR-33a-3p and hypoxia-inducible factor 1 alpha (HIF-1 alpha). Knockdown of LINC00518 expression sensitizes CMM cells to radiotherapy, suggesting a potential strategy to improve treatment outcomes.

Radiation-irritated skin and hyperpigmentation may impact the quality of life of breast cancer patients after whole breast radiotherapy

Chin-Nan Chu et al.

Summary: This study found that breast cancer patients after whole breast radiotherapy may experience skin dryness, hypersensitivity, and hyperpigmentation in the irradiated skin area. These radiation-induced skin lesions could lead to depressive psychological status and impact the quality of life in breast cancer patients after whole breast radiotherapy.

STING enhances cell death through regulation of reactive oxygen species and DNA damage

Thomas J. Hayman et al.

Summary: STING functions as an intrinsic regulator of tumor cell survival by controlling ROS homeostasis and affecting DNA damage and therapeutic resistance. Low STING expression is associated with worse tumor outcomes. Pharmacologic activation of STING enhances the effects of ionizing radiation, providing a rationale for therapeutic combinations of STING agonists and DNA-damaging agents.

Rapamycin Promotes ROS-Mediated Cell Death via Functional Inhibition of xCT Expression in Melanoma Under γ-Irradiation

Yunseo Woo et al.

Summary: The study investigated the effects of rapamycin on the interaction between tumor cells and macrophages, with a focus on regulating the mTOR pathway to improve tumor radioresistance. The results showed that rapamycin reduced secretion of IL-4 in tumor cells, decreased levels of YM1 in macrophages, and led to an increase in colocalized proportions of xCT and LC3 in tumor tissues. Overall, suppressing mTOR signaling in the tumor microenvironment may be beneficial for enhancing tumor response to radiotherapy.

Combination of radiotherapy and targeted therapy for melanoma brain metastases: a systematic review

Yi Ge et al.

Summary: Radiotherapy is an effective treatment for brain metastases from solid tumors, while immunotherapy has improved survival for metastatic cancer patients. Targeted therapy is a viable alternative for patients who cannot continue immunotherapy or have poor outcomes from it. The combination of radiotherapy and targeted therapy has theoretical support but limited data on efficacy and safety.

Stephanie Jonuscheit et al.

Summary: Niraparib and Talazoparib, as PARP 1/2 inhibitors, sensitize melanoma cells to ionizing radiation (IR), with healthy tissue being less affected. However, the heterogeneity of results suggests the need for prior testing of tumor cells for personalized treatment.

Justin C. Jagodinsky et al.

Summary: This study investigated the time course of STING-dependent IFN1 response following radiation in multiple murine tumor models. The results demonstrated the potential of TRT to stimulate IFN1 activation comparable to that observed with EBRT, which may be critical to the therapeutic integration of TRT with immunotherapies.
