Immune suppression in gliomas

Matthew M. Grabowski et al.

Summary: Gliomas evade immune detection and destruction through mechanisms such as dysfunctional T cells and NK cells, expansion of regulatory T cells and myeloid-derived suppressor cells, M2 phenotypic transformation in macrophages/microglia, upregulation of immunosuppressive factors, tumor microenvironment hypoxia, and iatrogenic effects of immunosuppressive treatments. Future research should focus on addressing these immunosuppressive mechanisms to develop treatment options with improved survival outcomes.

Concurrent Dexamethasone Limits the Clinical Benefit of Immune Checkpoint Blockade in Glioblastoma

J. Bryan Iorgulescu et al.

Summary: The study indicates that concurrent dexamethasone therapy may have detrimental effects on immunotherapeutic approaches in patients with glioblastoma, by reducing T-lymphocyte numbers and functionality, as well as decreasing myeloid and natural killer cell populations.

Inhibitory CD161 receptor identified in glioma-infiltrating T cells by single-cell analysis

Nathan D. Mathewson et al.

Summary: This study utilized single-cell RNA sequencing to uncover the gene expression and clonal landscape of T cells in diffuse gliomas, identifying potential effectors of anti-tumor immunity. The research revealed that KLRB1 and its associated transcriptional program play an inhibitory role in tumor immunity, and blocking its function enhances T cell killing of glioma cells.

Multifaceted Role of Matrix Metalloproteinases in Neurodegenerative Diseases: Pathophysiological and Therapeutic Perspectives

Tapan Behl et al.

Summary: Neurodegeneration is a pathological condition where the nervous system or neuron loses structure and function, with no known treatment yet, and matrix metalloproteinases (MMPs) play a crucial role in the progression of neurodegenerative diseases.

Circulating clonally expanded T cells reflect functions of tumor-infiltrating T cells

Liliana E. Lucca et al.

Summary: Understanding the relationship between tumor and peripheral immune environments could allow longitudinal immune monitoring in cancer. Analysis of T cells from both tumor and blood in patients with metastatic melanoma revealed that circulating T cells matching clonally expanded tumor-infiltrating T cells show gene signatures of effector functions, while features of exhaustion are predominantly displayed by tumor-exclusive T cells. Circulating TILs have unique transcriptional patterns that may be useful for interrogating T cell function in cancer immunotherapy.

Circulating Tumor Cells for Glioma

Huikai Zhang et al.

Summary: Liquid biopsy has been applied in various cancers but little progress has been made in validating circulating biomarkers for brain malignancies. Data on circulating tumor cells in glioma are limited, and their application as a biomarker has just begun.

EMP3 mediates glioblastoma-associated macrophage infiltration to drive T cell exclusion

Qun Chen et al.

Summary: The study found that EMP3 plays a critical role in immune modulation in GBM by affecting macrophage polarization and recruitment, leading to suppression of T cell infiltration into tumors and facilitating tumor progression. This suggests that EMP3 could be a potential therapeutic target for GBM treatment.

Platelets, immune cells and the coagulation cascade; friend or foe of the circulating tumour cell?

Mark P. Ward et al.

Summary: Circulating tumour cells (CTCs) are cancer cells that have unique characteristics allowing them to survive in the circulation, extravasate, and metastasize. Their interactions with components of the blood circulatory system influence their physical characteristics and metastatic capabilities, providing critical diagnostic and prognostic value in clinical settings. Platelets play a crucial role in the formation of metastatic niches and interact with CTCs to influence their fate, demonstrating potential for therapeutic disruption and targeted treatments in advanced cancers.

A single-cell map of intratumoral changes during anti-PD1 treatment of patients with breast cancer

Ayse Bassez et al.

Summary: The study found that in breast cancer patients, anti-PD1 treatment can lead to clonal expansion of PD1-expressing T cells in a subset of tumors, regardless of tumor subtype. The expansion mainly involves CD8(+) T cells and CD4(+) T cells with distinct immune cell characteristics.

TIGIT and PD-1 Immune Checkpoint Pathways Are Associated With Patient Outcome and Anti-Tumor Immunity in Glioblastoma

Itay Raphael et al.

Summary: Glioblastoma (GBM) remains a highly aggressive brain tumor with limited success in immune checkpoint (IC) inhibitors therapy. PD1 and TIGIT have been identified as top targets for GBM immunotherapy, and dual blockade of these molecules has shown improved survival in a murine GBM model. Additionally, this combination immunotherapy affects specific myeloid cells and restores T cell proliferation inhibited by immunosuppressive myeloid cells in human GBM tissue.
