Colchicine for prevention of vascular inflammation in Non-CardioEmbolic stroke (CONVINCE) - study protocol for a randomised controlled trial

Peter Kelly et al.

Summary: CONVINCE is a multi-center international Phase 3 clinical trial investigating the effect of low-dose colchicine on recurrent vascular events in patients with non-severe, non-cardioembolic stroke and TIA. The study will provide high-quality randomized data assessing the efficacy and safety of colchicine anti-inflammatory therapy for secondary prevention after stroke.

The use of blood biomarkers in precision medicine for the primary prevention of atherosclerotic cardiovascular disease: a review

Ty Sweeney et al.

Summary: Biomarkers are substances, structures, or processes that indicate the presence of a disease, infection, or environmental exposure. Studying blood biomarkers of ASCVD can improve the precision with which clinicians can diagnose and prevent the disease. Certain biomarkers have been highlighted as 'risk-enhancing factors,' aiding in shared decision-making.

Cardiac natriuretic peptides for diagnosis of covert atrial fibrillation after acute ischaemic stroke: a meta-analysis of diagnostic accuracy studies

Kejia Zhang et al.

Summary: Detection of atrial fibrillation (AF) after acute ischaemic stroke is crucial for preventing recurrence. In addition to heart rhythm monitoring, blood biomarkers such as B-type natriuretic peptide (BNP) and N-terminal pro-BNP (NT-proBNP) have been suggested as diagnostic tools for AF. While NT-proBNP shows better performance in diagnosing covert AF after acute ischaemic stroke compared to BNP, both biomarkers have limited utility as standalone decision-making tools.

Promising Biomarker Candidates for Cardioembolic Stroke Etiology. A Brief Narrative Review and Current Opinion

Arnold Markus et al.

Summary: Identifying the cause of stroke is crucial in guiding personalized prevention strategies, and biomarkers associated with etiology could play a key role in this. As personalized medicine advances, researchers are increasingly focusing on etiology-related biomarkers to gain deeper insight into specific stroke mechanisms. Validation of promising biomarkers and their implementation in clinical trials are necessary to further understand stroke pathophysiology and improve prevention strategies.

Anna Dieden et al.

Summary: In this observational study, a multiplex proteomic platform was used to explore associations between 92 targeted proteins and deteriorating vascular health phenotypes in a cohort of black and white African school teachers. The study revealed significant associations between hypertension, carotid intima-media thickness (cIMT), pulse wave velocity (PWV) and specific proteins related to inflammation, chemotaxis, coagulation or proteolysis. Additionally, ethnicity played a role in some of these associations, highlighting the complexity of the relationships between proteins and vascular health markers.
