Salmonella enterica Serovar Typhimurium Exploits Cycling through Epithelial Cells To Colonize Human and Murine Enteroids

Petra Geiser et al.

Summary: This study reveals the complete infection cycle of Salmonella enterica serovar Typhimurium within human and mouse enteroids, and demonstrates how bacterial invasion of the epithelium strongly fuels expansion in the luminal compartment through a mechanism involving the death and expulsion of bacterium-infected epithelial cells.

Regulatory protein HilD stimulates Salmonella Typhimurium invasiveness by promoting smooth swimming via the methyl-accepting chemotaxis protein McpC

Kendal G. Cooper et al.

Summary: The protein HilD of Salmonella Typhimurium coordinates motility and host cell invasion by upregulating flagellar genes and a secretion system. In addition, HilD also modulates swimming behavior by upregulating a subunit of the chemotactic receptor array, which is important for invasion of epithelial cells.

The Salmonella effector protein SopD targets Rab8 to positively and negatively modulate the inflammatory response

Huan Lian et al.

Summary: The Salmonella effector protein SopD can both activate and inhibit the host inflammatory response by targeting the Rab8 GTPase. By modulating Rab8, SopD functions to counteract and stimulate inflammation, ultimately demonstrating the evolution of a bacterial effector protein with dual activities towards the same host cellular target to regulate the inflammatory response.

Epithelium-autonomous NAIP/NLRC4 prevents TNF-driven inflammatory destruction of the gut epithelial barrier in Salmonella-infected mice

Stefan A. Fattinger et al.

Summary: The gut epithelium serves as a critical barrier against infection by Gram-negative bacteria, with the NAIP/NLRC4 inflammasome playing a key role in sensing and responding to such infections. In NAIP/NLRC4-deficient mice, severe enteropathy and epithelial barrier breakdown were observed, while hosts with functional NAIP/NLRC4 were able to control bacterial loads through a heterogeneous enterocyte death response, preventing excessive inflammatory reactions. These findings highlight the importance of the NAIP/NLRC4 inflammasome in maintaining gut homeostasis during bacterial infections.

Bacterial detection by NAIP/NLRC4 elicits prompt contractions of intestinal epithelial cell layers

Pilar Samperio Ventayol et al.

Summary: The study found that infection with Salmonella Typhimurium triggers focal contractions in intestinal epithelial cells, which are initiated by the NAIP/NLRC4 inflammasome and can prevent infected cell death and expulsion, thereby protecting the integrity of the intestinal epithelium.

Salmonella enterica Serovar Typhimurium SPI-1 and SPI-2 Shape the Global Transcriptional Landscape in a Human Intestinal Organoid Model System

Anna-Lisa E. Lawrence et al.

Summary: The human intestinal organoid (HIO) model provides a more accurate platform to study host transcriptional responses to foodborne pathogens like Salmonella enterica Typhimurium. Infection with S. Typhimurium strongly stimulates proinflammatory gene expression and downregulates genes associated with cell cycle and DNA repair, leading to reduced cellular proliferation. The findings highlight the importance of Salmonella pathogenicity island-1 (SPI-1) and SPI-2-encoded type 3 secretion systems in modulating early host responses during enteric infection.

Investigation of the invasion mechanism mediated by the outer membrane protein PagN of Salmonella Typhimurium

Emilie Barilleau et al.

Summary: PagN, a well-conserved protein among different species and subspecies of Salmonella, interacts with heparan sulfate proteoglycan (HSPG) to mediate invasion, but this interaction alone is not sufficient for internalization mechanism. Instead, PagN induces a phosphatidylinositol 3-kinase signaling pathway, leading to a Zipper-like entry mechanism similar to the Salmonella outer membrane protein Rck.
