Comparative metagenomic discovery of the dynamic cellulose-degrading process from a synergistic cellulolytic microbiota

Ming Yang et al.

Summary: In this study, a microbiota (FPDM) with cellulose-degrading ability was cultivated to investigate the dynamic process of cellulose biodegradation and explore potential cellulases. The results showed that Sporocytophaga and Cohnella dynamically dominated the synergistic degradation of cellulose in different periods, while unannotated genes were speculated to participate in cellulose catabolism. The dynamic cellulose-degrading pathway of FPDM was predicted based on changes in hydrolysates, community structure, and gene abundance, providing a new perspective for identifying key cellulolytic strains and enzymes.

Degradation of plastic waste using stimulated and naturally occurring microbial strains

Navid Taghavi et al.

Summary: This study investigated the biodegradation capability of microbial strains derived from soil, activated sludge, farm sludge, and worms' excreta on high-density polyethylene, polystyrene foam, polypropylene, and polyethylene terephthalate under unstimulated and stimulated conditions. The most effective plastic-degrading microbes were identified as Penicillium raperi, Aspergillus flavus, Penicillium glaucoroseum, and Pseudomonas sp. In the mixed condition, incubation of polyethylene with Aspergillus flavus showed the highest weight loss at 5.5%. Further analysis using Fourier Transform Infrared Spectroscopy revealed the formation of new functional groups on the treated plastics.

Summary: The importance of understanding the roles played by different microbial communities and their symbiotic associations in facilitating the breakdown of organic substrates in methane production has been highlighted. Traditional approaches have limitations in elucidating the actual roles of microorganisms within communities, prompting the introduction of metagenomics coupled with metabolomics for a more intricate understanding of biological processes in anaerobic digestion systems.

An extracellular lipase from Amycolatopsis mediterannei is a cutinase with plastic degrading activity

Yeqi Tan et al.

Summary: An extracellular lipase from Amycolatopsis mediteranei (AML) was cloned and studied for potential applications in process biotechnology, with its 3D structure elucidated by comparative modelling. AML was found to be a cutinase with specificity for medium chain fatty acyl moieties and capable of rapidly hydrolyzing certain aliphatic plastics under mild conditions. The findings suggest that minor changes in the active site region of AML can significantly affect substrate preference, expanding the potential applications for bioremediation of plastics.

Genomic and functional analyses of fungal and bacterial consortia that enable lignocellulose breakdown in goat gut microbiomes

Xuefeng Peng et al.

Summary: The study found that microbial consortia dominated by anaerobic fungi outperformed bacterially dominated consortia in methane production and cellulose degradation. Cross-domain partnerships between fungi and methanogens enabled the production of acetate, formate, and methane, while bacterially dominated consortia mainly produced short-chain fatty acids. The division of labor among herbivore anaerobes for plant biomass degradation could be utilized for industrial bioprocessing.

Single strain control of microbial consortia

Alex J. H. Fedorec et al.

Summary: The scope of bioengineering is expanding to include the design of microbial communities, and researchers have shown that a stable two-strain community can be achieved by engineering a strain of Escherichia coli to secrete a toxin in response to competition. Experimental and mathematical evidence demonstrates that this system can produce stable populations with tunable compositions controlled by easily adjustable parameters. This approach allows for the creation of tunable, stable two-strain consortia with the potential to interface with natural microbiomes.

Methanogenic archaea use a bacteria-like methyltransferase system to demethoxylate aromatic compounds

Julia M. Kurth et al.

Summary: The study identified a novel methoxydotrophic archaeon capable of converting methoxylated aromatic compounds to methane. Transcriptomics and proteomics analysis revealed that the archaeon used an O-demethylation/methyltransferase system similar to acetogenic bacteria. Enzymatic assays indicated a two-step mechanism in which the methyl group is transferred from the methoxy compound to cobalamin and then further transferred to tetrahydromethanopterin.

Summary: This study demonstrated the potential of white rot fungi Pycnoporus sanguineus in biodegrading triphenyl phosphate (TPhP) and identified the degradation pathway involving oxidative cleavage, hydroxylation, and methylation. The biotreatment by P. sanguineus significantly reduced the biotoxicity of TPhP, and its cooperation with indigenous degraders promoted efficient TPhP biodegradation in the water-sediment system. This research provided insights into the mechanisms, pathways, and risk assessment of TPhP biodegradation by P. sanguineus, confirming the feasibility of using this fungus for TPhP bioremediation applications.

Mixed Acid Fermentation of Carbohydrate-Rich Dairy Manure Hydrolysate

Abel T. Ingle et al.

Summary: The study aimed to investigate the recovery of carbon from dairy manure fibers as medium-chain fatty acids (MCFAs) through various pretreatments. The decrystalization dilute acid pretreatment (DCDA) was found to be the most effective, producing carbohydrate-rich hydrolysates suitable for fermentation. Bioreactors fed with DCDA hydrolysates produced acetic acid and butyric acid as major end products, with transient production of MCFAs. Microbial community analysis revealed the involvement of lactic acid bacteria, Megasphaera, and Caproiciproducens in MCFA and C4 production.
