Inherited GATA2 Deficiency Is Dominant by Haploinsufficiency and Displays Incomplete Clinical Penetrance

Carmen Oleaga-Quintas et al.

Summary: Germline heterozygous mutations of GATA2 can lead to various hematological and clinical phenotypes, including mycobacterial diseases. Clinical penetrance for GATA2 deficiency-related manifestations such as mycobacterial diseases was found to be incomplete, with affected relatives spanning a wide range of ages, emphasizing the importance of genetic testing for patients at any age.

Human ancient DNA analyses reveal the high burden of tuberculosis in Europeans over the last 2,000 years

Gaspard Kerner et al.

Summary: This study revealed the history of human exposure to tuberculosis in Europe and found that the TYK2 P1104A variant originated from common ancestors of West Eurasians around 30,000 years ago. The frequency of the variant fluctuated significantly over the past 10,000 years, with a dramatic decrease after the Bronze Age. The drop in frequency was attributed to strong negative selection starting around 2,000 years ago, resulting in a 20% relative fitness reduction in homozygotes, indicating a heavy burden of tuberculosis on European health over the last two millennia.

The Ever-Increasing Array of Novel Inborn Errors of Immunity: an Interim Update by the IUIS Committee

Stuart G. Tangye et al.

Summary: The latest classification of inborn errors of immunity/primary immunodeficiencies by the International Union of Immunological Societies Expert Committee, published in January 2020, quickly became outdated due to the rapid pace of genetic variant discoveries in the field. To bridge the gap until the next official report in 2022, researchers have highlighted 26 additional monogenic gene defects as novel causes of immune defects.

Inherited deficiency of stress granule ZNFX1 in patients with monocytosis and mycobacterial disease

Tom Le Voyer et al.

Summary: The study reports four patients with intermittent monocytosis and mycobacterial disease, homozygous for ZNFX1 variants predicted to be loss of function. Despite lacking known inborn errors of IFN-gamma, the patients show normal responses to IFN-gamma but have defective ZNFX1 protein, essential for monocyte homeostasis and protective immunity to mycobacteria.

Inherited PD-1 deficiency underlies tuberculosis and autoimmunity in a child

Masato Ogishi et al.

Summary: A study on a patient with a homozygous loss-of-function mutation in PDCD1 reveals dysregulated immune features involving IL-6, IL-23, STAT3, and ROR gamma T, which may be potential targets for the treatment of PD-1 blockade-induced autoimmunity. The research highlights the essential role of human PD-1 in governing antimycobacterial immunity and self-tolerance, while identifying actionable molecular targets for the diagnostic and therapeutic management of TB and autoimmunity in patients on PD-1 blockade.
