Bioengineered Skeletal Muscle as a Model of Muscle Aging and Regeneration

Nika Rajabian et al.

Summary: This study developed a three-dimensional bioengineered tissue model mimicking aged muscle, demonstrating the characteristics of senescent muscle and the inability of senescent SkM tissues to regenerate. This model may serve as a powerful platform for studying aging-related issues.

Engineering skeletal muscle tissues with advanced maturity improves synapse formation with human induced pluripotent stem cell-derived motor neurons

Jeffrey W. Santoso et al.

Summary: In order to develop effective treatments for neuromuscular diseases, human-relevant in vitro models of neuromuscular tissues are essential. This study improved neuromuscular junctions formed by human induced pluripotent stem cell (hiPSC)-derived motor neurons through optimizing the maturity of the co-cultured muscle tissue, specifically using engineered chick muscle tissues. The engineered chick muscle tissues displayed increasing maturity in NMJs, revealing new insights into the molecular pathways involved in enhancing NMJ formation.

Macrophages provide a transient muscle stem cell niche via NAMPT secretion

Dhanushika Ratnayake et al.

Summary: Through muscle injury models in zebrafish, it has been found that specific macrophage populations 'dwell' within the injury site, providing a transient niche for stem cell proliferation. The signals released by these macrophages directly stimulate stem cell proliferation, governing the repair process.

Myogenesis modelled by human pluripotent stem cells: a multi-omic study of Duchenne myopathy early onset

Virginie Mournetas et al.

Summary: This study utilized cells from DMD patients and induced pluripotent stem cells to investigate the pathogenesis of DMD through multi-omic analysis, revealing that pathological changes of DMD start as early as the embryonic stage, suggesting a need to reconsider the role of dystrophin in muscle development.

Summary: The study developed a 3D in vitro model of human skeletal muscle that recapitulates the pathophysiology of Pompe disease. They identified a Pompe disease-specific transcriptional signature and observed a partial reversal of the signature upon in vitro treatment of myobundles with rhGAA.

De novo revertant fiber formation and therapy testing in a 3D culture model of Duchenne muscular dystrophy skeletal muscle

Majid Ebrahimi et al.

Summary: The study demonstrates that 3D human skeletal muscle microtissues (hMMTs) offer a valuable model to investigate DMD pathology and potential therapeutic strategies. This system allows for the observation of DMD-associated phenotypes not seen in traditional 2D cultures, providing a platform for pre-clinical research and drug testing.

A muscle fatigue-like contractile decline was recapitulated using skeletal myotubes from Duchenne muscular dystrophy patient-derived iPSCs

Tomoya Uchimura et al.

Summary: This study demonstrates the development of a disease model using MYOD1-induced differentiation of hiPSCs into functional skeletal myotubes, which recapitulates declines in contractile performance in dystrophic myotubes through long-term EFS training. Screening of clinically relevant drugs using this model identifies three compounds that ameliorate this decline, and the feasibility of adapting the model to a 96-well culture system using optogenetic technology for large-scale screening is validated.
