Summary: Despite advancements in chemical recycling of synthetic polymers, efficient conversion of post-consumer PET into TPA under ambient conditions remains a challenge. Mechanochemistry and solid-state ball milling show promise in achieving this conversion, as demonstrated in this study on a gram-scale depolymerization of PET into TPA.

Engineering Escherichia coli for the utilization of ethylene glycol

Summary: In this study, ethylene glycol (EG) is explored as a novel feedstock for producing chemicals and fuels. By engineering Escherichia coli (E. coli) to consume EG, with glycolate production as a case study, oxygen concentration is identified as a crucial factor in the assimilation and use of EG as a substrate. Two oxygen-based strategies are developed and tested in fed-batch bioreactors to optimize glycolate production, with promising results demonstrating the potential of EG as a renewable starting material for commercial biosynthesis.

A new-to-nature carboxylation module to improve natural and synthetic CO2 fixation

Summary: The development of a new enzyme GCC with improved catalytic efficiency is a key step towards enhancing carbon efficiency in various processes. By combining active-site design and engineering of other enzymes, a carboxylation module was created, which can be interfaced with natural metabolism and CO2 fixation, providing new opportunities for biotechnology and agriculture.

Constructing an ethanol utilization pathway in Escherichia coli to produce acetyl-CoA derived compounds

Summary: By engineering microbes, a two-step heterologous ethanol utilization pathway was designed and constructed in Escherichia coli to convert ethanol into acetyl-CoA efficiently without ATP consumption. The optimized pathway generated two molecules of NADH per molecule of ethanol. The engineered E. coli strain successfully consumed approximately 8 g/L of ethanol as the sole carbon source, and produced 1.1 g/L of polyhydroxybutyrate in combination with the biosynthetic pathway within 96 h. Moreover, the feasibility of converting ethanol into different classes of acetyl-CoA derived compounds was demonstrated by producing prenol in an engineered E. coli strain.
