Lapachol in the Design of a New Ruthenium(II)-Diphosphine Complex as a Promising Anticancer Metallodrug

Luminescent naphthalimide-tagged ruthenium(ii)-arene complexes: cellular imaging, photocytotoxicity and transferrin binding

Summary: Two water-soluble piano-stool shaped ruthenium(ii)-arene complexes were designed as emissive photocytotoxic agents tagged with morpholine as the lysosome targeting moiety. They showed enhanced photo-cytotoxicity under low-power blue LED light irradiation, preferentially localized in lysosomes and nuclei, effectively targeting and damaging nuclear DNA for photodynamic therapy. Due to enhanced lipophilicity of one complex, it showed higher internalization into cancer cells, with possible binding through histidine residues and internalization via endocytosis. Luminescence properties were utilized to study cellular uptake mechanisms via endocytosis.
