Isolation, Structure Elucidation, Semi-Synthesis, and Structural Modification of C19-Diterpenoid Alkaloids from Aconitum apetalum and Their Neuroprotective Activities

Summary: Five new aconitine-type C-19-diterpenoid alkaloids, apetalrines A-E (1-5), were isolated from Aconitum apetalum. Among them, apetalrine B (2) exhibited good neuroprotective effects by inhibiting cell apoptosis, confirmed through a series of studies including flow cytometry, staining, and Western blotting. Further synthesis and evaluation of derivatives of compound 2 revealed its potential for neuroprotective applications with low cytotoxicity.

Limonin as a Starting Point for the Construction of Compounds with High Scaffold Diversity

The reactions involved in constructing these compounds include novel ring cleavage, rearrangements, and cyclizations, resulting in the discovery of new reactions such as B-ring cleavage and B/C-ring rearrangement. Computational analysis revealed the production of 52 different scaffolds/ring systems, with 36 being unprecedented. Phenotypic screening identified compounds with activity against a panel of cancer cell lines.
