Cryo-EM structures of intermediates suggest an alternative catalytic reaction cycle for cytochrome c oxidase

F. Kolbe et al.

Summary: Cytochrome c oxidase is a fundamental enzyme in life, and the authors present cryo-EM structures of different intermediate states that suggest an alternative reaction cycle. The enzyme uses electrons from cytochrome c molecules to reduce oxygen to water, with structures showing peroxide dianion and intact dioxygen molecule in different states. This suggests a need for a 180-degree turn in the enzyme's catalytic cycle.

The Redox-Active Tyrosine Is Essential for Proton Pumping in Cytochrome c Oxidase

Margareta R. A. Blomberg

Summary: Cellular respiration involves electron transport via enzyme complexes to terminal Cytochrome c oxidase, where molecular oxygen is reduced to water, establishing a transmembrane electrochemical gradient using the free energy released. The redox-active tyrosine in CcOs plays a crucial role in energy conservation by allowing proton pumping in all reduction steps. The unprotonated tyrosine provides a mechanism for coupling the uptake of two protons per electron in every reduction step, ensuring secure proton pumping.
