Chiral Cyclopentadienyl Ligands: Design, Syntheses, and Applications in Asymmetric Catalysis

Summary: The development of new chiral ligands is crucial for stereocontrol in metal-catalyzed reactions, especially with the increasing demand for bioactive molecules as single enantiomers. The successful application of Cp-X ligands in metal-catalyzed transformations allows access to valuable chiral molecules, with critical comparisons of existing ligand families and discussion of future research directions for further enhancing their performance and application in enantioselective catalysis.

Recent Advances in Enantioselective Direct C-H Addition to Carbonyls and Michael Acceptors

Different types of chiral catalysts can achieve enantioselective C-H addition reactions with various substrates, providing a direct access to valuable chiral fragments. The reaction mechanisms of these enantioselective C-H additions are also introduced in this review.

Asymmetric hetero-Michael addition to α,β-unsaturated carboxylic acids using thioureaeboronic acid hybrid catalysts

Summary: This review summarizes the first catalytic asymmetric Michael addition of heteroatomic nucleophiles to alpha,beta-unsaturated carboxylic acids. Hybrid catalysts are designed to activate the unsaturated carboxylic acids, leading to synthetic applications to biologically active compounds. The mechanistic consideration of multicomponent borate complexes is also discussed in this work.

Summary: A novel method has been developed in this study for controlling carbonyl alpha-chirality and functionalizing beta-position through the conjugate addition-asymmetric protonation (CAAP) of alpha,beta-unsaturated carboxylic acids using chiral thiourea-amino boronic acid hybrid catalysts. This method is also applied in the asymmetric synthesis of biologically active compounds.

Siloxy Esters as Traceless Activators of Carboxylic Acids: Boron-Catalyzed Chemoselective Asymmetric Aldol Reaction**

Summary: This study identified that the in situ pre-conversion of carboxylic acids to siloxy esters facilitated the catalytic asymmetric aldol reaction of multifunctional substrates. A combination of experimental and computational studies rationalized the reaction mechanism and proposed Si/B enediolates as the active species. Silyl ester formation promoted enolization and catalyst turnover by acidifying the alpha-proton of substrates and attenuating poisonous Lewis bases to the boron catalyst.

Visible-Light-Driven α-Allylation of Carboxylic Acids

Summary: A boron-catalyzed alpha-allylation of carboxylic acids with allylsulfones under blue LED irradiation conditions has been developed. The photoexcitation of catalytically generated boron enediolates induces single-electron transfer to the allylsulfones, leading to subsequent C-C bond formation and the construction of sterically demanding quaternary carbon centers at the position a to the carboxy group. The p-extended ligand on the boron atom is likely crucial for the efficient photoexcitation of the boron enediolates.
