Navigating the Pauson-Khand Reaction in Total Syntheses of Complex Natural Products

Zhen Yang

Summary: The total synthesis endeavors contribute to the discovery and invention of new synthetic reactions to advance organic synthesis. Bioactive compounds share common features, such as molecular complexity, protein-binding ability, structural rigidity, and three-dimensionality. Total synthesis offers an alternative solution for generating scarce natural products and their derivatives for studying their biological functions.

Unprecedented Reactivity of γ-Amino Cyclopentenone Enables Diversity-Oriented Access to Functionalized Indoles and Indole-Annulated Ring Structures

Chenna Jagadeesh et al.

Summary: An unexpected Lewis acid promoted displacement of the latent reactive gamma-amino group on cyclopentenone was discovered in this study, providing an unparalleled opportunity for enone functionalization and annulations with indole derivatives. A wide range of C3/N-indolyl enones and indole alkaloid-like compounds were accessed in excellent yields and selectivity through a one-pot operation, with the influence of the gem-diaryl group appearing crucial in the mechanism involving a Piancatelli-type rearrangement.

Enantioselective Silicon-Directed Nazarov Cyclization

Jin Cao et al.

Summary: This study presents highly enantioselective silicon-directed Nazarov reactions cooperatively catalyzed by a Lewis acid and a chiral Bronsted acid, which allows for the synthesis of chiral cyclopentenones with diverse substituent patterns not achievable by other catalytic enantioselective reactions. The positioning of the double bond in the product is determined by the stabilizing effect of the silicon group in the dienone substrate on the beta-carbocation intermediate, and the enantioselectivity of the reaction is attributed to a chiral Bronsted acid promoted proton transfer reaction of the enol intermediate.

Catalytic Asymmetric Radical-Mediated Three-Component Piancatelli-Type Rearrangement of Furylalkenes

Lei Xu et al.

Summary: This study developed a new asymmetric radical-mediated Piancatelli-type rearrangement reaction, which allows for a three-component reaction of diverse carbon-centered radical precursors and anilines with high yields, enantioselectivities, and diastereoselectivities. The synthesized products can be applied for late-stage functionalization of valuable drug targets or natural products. The diastereoselectivity of the rearrangement reaction is controlled by Dy(III)-accelerated Z/E isomerization of Piancatelli intermediates, and the chiral Bronsted acid-catalyzed 4π-electrocyclization of pentadienyl carbocation intermediates is a key step determining both reaction rate and enantioselectivity.
