Revealing the Viscosity Changes in Lipid Droplets during Ferroptosis by the Real-Time and In Situ Near-Infrared Imaging

Summary: The study revealed the changes of lipid droplets viscosity during ferroptosis using a novel viscosity-sensitive near-infrared fluorescent probe. The experiment results showed a significant increase in lipid droplets viscosity during ferroptosis, while there was nearly no change in cells treated with ferroptosis inhibitors simultaneously.

Intramolecular Spirocyclization Enables Design of a Single Fluorescent Probe for Monitoring the Interplay between Mitochondria and Lipid Droplets

Summary: A novel fluorescent probe MT-LD was designed in this study to label lipid droplets and mitochondria, allowing for the visualization of their interaction during processes such as oleic acid treatment and starvation-induced autophagy. The probe revealed an increase in mitochondria-LD contact and peridroplet mitochondria amount in different tissues, such as brown adipose tissue and lung tissue. This work provides a valuable tool for investigating the interplay between mitochondria and lipid droplets, promoting research in fatty acid metabolism and autophagy.

Simultaneous visualization of lipid droplets and lysosomes using a single fluorescent probe

Summary: The study developed a polarity-sensitive naphthalimide derivative fluorescent probe LD-Lys with high photostability and low cytotoxicity, which can simultaneously image LDs and lysosomes by showing strong and weak fluorescence, respectively. The dual-color imaging of LDs with bright yellow emission and lysosomes with weak red emission was achieved using a confocal laser scanning microscope. By monitoring the location changes, real-time tracking of LDs and lysosomes in cells and visualizing the size, morphology, and distribution of LDs and lysosomes in different tissues were feasible with LD-Lys, making it a promising fluorescent agent for studying the LD-lysosome interplay and metabolism diseases.

This probe enables the highest resolution imaging of LDs to date, time-lapse live imaging, visualization of the fusion process of nascent LDs, and high-quality 3D imaging, showcasing its utility in STED super-resolution imaging.
